Backgrounds and ObjectiveGastric cancer originates in gastric mucosa epithelial cells, all can happen in anypart of the stomach. Worldwide, the stomach cancer incidence is in the fourth, andsecond tumor mortality. Every year around1000,000new cases of gastric carcinomahappen, and death of about800,000, China is one of the countries that have highmorbidity and mortality of gastric cancer. Cancer stem cell theory in recent years ismore and more taken attention by people, the theory is that tumor stem cells areprimary tumor tissue in a class of self-renewal, unlimited proliferation andmulti-directional differentiation potential of cells, and are the root of tumor growth,metastasis and recurrence. This theory provides a new direction to the research oftumor. Wnt signaling pathways involved in stem cell signaling network, is associatedwith the occurrence and development of tumor. As target genes of Wnt signalingpathways, G protein coupled receptor Lgr5mainly expresses in muscle, placenta,spinal cord, in the brain tissue. In recent years, the study found that Lgr5as one of thecancer stem cell markers, expresses in a wide variety of tumor tissues, prompt Lgr5has certain correlation with the incidence of tumor. Wnt1is known to the firstmember of the19human Wnt genes family, both at home and abroad, the studyfound the Wnt1abnormalities, ectopic expression is involved in many biologicalprocesses, such as cell differentiation, cell polarity formation, cell migration andproliferation, and is closely related to the multiple system of human tumor. At present,the domestic and foreign reports of Lgr5, Wnt1joint research in gastric cancer is notvery much. This study by immunohistochemical method has combined detection ingastric cancer tissues and corresponding expression in normal tissue adjacent to carcinoma and explore the role of both in gastric cancer development and the mutualrelationship, which provides much theoretical basis for the clinical diagnosis andprognosis and treatment.MethodsThe expression of Lgr5and Wnt1was detected by immunohistochemistry in76cases of gastric cancer and30cases of normal tissues nearby the cancer. Theassociation between Lgr5and Wnt1protein expression with clinicopathologicalparamters was analyzed. We used SPSS17.0software to analyze database, and usedvariability chi-square test to analyze the express of Lgr5and Wnt1in the two tissues,and then used spearman correlation analysis which is used to analyze the relationbetween Lgr5and Wnt1in gastric cancer and used a=o.o5as the inspection level.ResultsThe positive rate of Lgr5and Wnt1expression in gastric cancer wassignificantly higher than those in normal tissues nearby the cancer(P <0.05). Theexpression of Lgr5and Wnt1had nothing with the age and sex,but was correlatedwith the size, TNM’s stage, histopathological grade and lymph node metastasis (all P<0.05). There was a positive correlation between the expressions of Lgr5and that ofWnt1in gastric cancer (r=0.504, P <0.01).ConclusionLgr5and Wnt1might play important roles in the progression, differentiation,infiltration, and metastasis of gastric cancer, and might be the indicators for diagnosisof gastric cancer, they also may be used as early diagnostic markers and therapeutictargets for gastric cancer.
