ObjectiveTo detect papillary thyroid carcinoma FHIT gene promoter methylation statusand its impact on mRNA and protein expression, to explore the meaning of the FHIToccurs and development in papillary thyroid carcinoma.MethodsUsing methylation-specific polymerase chain reaction (Methylation-specificPCR, MSP) to detect60cases of papillary thyroid carcinoma (PTC) and theircorresponding adjacent tissues (NCE) in the FHIT gene promoter methylation statususing rt-PCR to detect the expression of FHIT gene mRNA, using Western blottechnique (Western-Blot) detecting the cases of FHIT protein expression level ofpapillary thyroid carcinoma and the corresponding normal thyroid tissue.ResultsThere is no case with FHIT gene promoter methylation in NCE, but38.3%(23/60) methylation occurs in PTC tissues, and its gene methylation was positivelycorrelated with incidence of pathological grade, TNM stage and lymph nodemetastasis (P <0.05); while NCE and PTC, the positive rate of FHIT proteinexpression was100%(70/70) and41.7%(25/60) respectively, and FHIT proteinexpression was negatively correlated with its pathological grade, TNM stage andlymph node metastasis (P <0.05). The FHIT gene expression in PTC was45.0%,significantly lower than the NCE, PTC mRNA expression of FHIT gene in patientswith pathological grade, TNM stage and lymph node metastasis were correlated, the difference was statistically significant (P <0.05).ConclusionFHIT gene promoter methylation is likely to be an important mechanism of geneinactivation, and may plays an important role of the occurrence and development inPTC.
