Objective: In recent decades, the incidence of thyroid tumor weresignificantly high all of the world, in the endocrine system of malignant tumor,thyroid cancer has risen to the first place, thyroid papillary carcinoma(papillary thyroid carcinoma, PTC) is one of the most common pathologicaltypes.PTC’s main treatment including surgery and endocrine therapy, andhormone therapy did not fully support of experimental data, the purpose ofthis paper is to explore PTC cell membrane thyroid stimulating hormonereceptor (TSHR) methylation level, so as to individual endocrine therapypostoperatively in patients with thyroid papillary carcinoma.Methods:Collected surgical resection of the thyroid tissues of41casesfrom the second hospital, Hebei Medical University,east campus general2department from May2013to July2013.22cases of Thyroid papillarycarcinoma,9cases of thyroid benign lesions and10cases of thyroid tissueadjacent to carcinoma.Using real-time quantitative PCR technique to detect10cases of thyroid papillary carcinoma,10normal tissue adjacent to carcinomaand9cases of thyroid benign lesions the contents of TSHR gene.Bymethylation specific PCR (MSP) technology to detect TSHR genemethylation positive rate in three groups.Application of SPSS13.0statisticalsoftware one-way ANOVA analysis of variance, chi-square statistic methodand Fisher’s exact probability method to analyze the data. Inspection standardfor P <0.05was statistically significant.Results:Measurement data to mean±standard deviation. The gene ofTSHR content in10cases of thyroid papillary carcinoma tissues is7.132±1.605.The gene of TSHR content in9cases of thyroid benign lesionsis6.016±2.650.The gene of TSHR content in10cases of normal thyroidtissue adjacent to carcinoma is6.396±1.014. P=0.416. Can be thought of content of TSHR gene in papillary thyroid carcinoma, thyroid benign lesionsand normal tissue adjacent to carcinoma have no difference.The methylation rate of TSHR gene in PTC is54.55%(12/22); Themethylation rate of TSHR gene in thyroid benign lesions is11.11%(1/9);Themethylation rate of TSHR gene in normal thyroid tissue adjacent to carcinomais10.00%(1/10).And the rate of methylation of TSHR gene in PTC wassignificantly higher than of thyroid benign lesions and normal thyroid tissueadjacent to carcinoma.TSHR methylation rate in thyroid papillary carcinomais higher than in thyroid benign lesions and normal thyroid tissue.Fisher’sexact probability method for the further application of thyroid papillarycarcinoma, thyroid benign lesions and normal thyroid tissue adjacent tocarcinoma are compared, and two of PTC with thyroid benign lesions, PTCwith normal thyroid tissue adjacent to carcinoma difference was statisticallysignificant, can be thought of in PTC with thyroid benign lesion group, normalthyroid tissue adjacent to carcinoma TSHR gene methylation rate(P=0.045and P=0.024. Between thyroid benign lesions and normal thyroid tissueadjacent to carcinoma,the methylation positive was not different.(P=1.000).According to age, sex, tumor size, tumor number, whether lymph nodemetastasis, whether distant metastasis, TNM staging and thyroid functiongroup respectively.The methylation rate of cases of tumor≤10mm was12.50%(1/8), which was higher the methylation rate of cases of tumor>10mm(78.57%11/14).The methylation rate of cases of multiple tumors was87.50%(7/8), which was higher than the methylation rate of cases of singletumo(r35.71%;5/14).Using Fisher’s exact probability method to analysis.Thedifference was statistically significant(P=0.006and P=0.031).According tothe grope of tumor size and tumor number, the gene of TSHR methylationpositive difference was statistically difference, can be thought of tumor sizeand tumor number had something to do with the methylation of TSHR gene.But age,gender, whether lymph node metastasis, whether distant metastasisand TNM stages had nothing to do with TSHR gene methylation positive rate. Conclusion:1Content of TSHR gene in papillary thyroid carcinoma, thyroid benignlesions and normal tissue adjacent to carcinoma have no difference.2TSHR methylation rate in thyroid papillary carcinoma is higher than therate in thyroid benign lesions and normal thyroid tissue.3TSHR gene methylation is only related to tumor size and number, and hasnothing to do with the other clinical parameters.4TSHR gene methylation detection can be used for individualizedendocrine therapy postoperatively in patients with thyroid papillarycarcinoma.
