ObjectiveTo investigate the effects of aminoguanidine and baicalein on cerebrovascularremodeling in a rat model of diabetes mellitus and the possible mechanism byobserved ultrastructures of vessels and the expression change and the possiblemechanism of CD34and EGFL-7in rat brain tissue.Method48SD rats were randomly divided into following groups: aminoguanidine(AG)group(group A), Baicalein group(group B), diabetic group(group D) and controlgroup(group C).Group C was received pelleted food,group A,B,D were made byusing high fat and sugar diet in combination with small dose of intraperitonealinjection of streptozotocin, and then group A and group B was givenaminoguanidine,baicalein150mg/(kg.d) through oral gavaging, group D and C weregiven equivalent saline through oral gavaging. ultrastructures of vessels andperivascular and the expression of hematopoietic progenitor cell antigen CD34,Human Epidermal growth factor Like Domain Protein, Multiple7(EGFL-7) in brain tissue were determined after16weeks.Result1. under a transmisson electron microscope, group C was normal. Group Dshowed vascular stenosis, deformation, a large number of perivascular edema,basement membrane structure is fuzzy.Group A and B showed much better than thesein the group D. A comparison of group A and B showed no significant differences.2. By immunohistochemistry the expression of CD34in the group A,B and Dwere significantly lower than that in group C(P<0.05),and that in the group A and Bwere increased compared with group D(P <0.05), As compared with group A, groupB was no significant difference(P>0.05)3. By immunohistochemistry the EGFL-7expression in the group A,B,D weresignificantly increased compared with the control group(P<0.05),and that in the groupA,B were decreased compared with group D(P<0.05). EGFL-7expression in group Aand group B were no significantly difference(P>0.05).conclusionsThere are changs of cerebrovascular remodeling remodeling in diabetic rats.Aminoguandine and Baicalein can restrain cerebral vascular remodeling and protectblood vessels and nervous.
