Objective:To study the expression levels and diagnostic value of miRNA-21and miRNA-126as a biomarker of malignant pleural effusion in patients of non-small cell lung cancerMethods:We collected the pleural effusion of50NSCLC patients with malignant and benign pleural effusion in the respiratory medicine of the third hospital of changsha and the second Xiangya hospital of central south university from Septemper2012to April2013. We measured the expression of miR-21and miR126using real-time PCR in the samples of pleural effusion. Mann-whitney U test was used to compare the expression difference of miR-21and miR-126between malignant and benign pleural effusion. And ROC was used to evaluate the diagnostic value of NSCLC with malignant pleural effusion by the expression level of miR-21、miR-126.Results:(1) MiRNAs were extractd using Trizol. The expression levels of miR-21and miR-126were quantified by real-time qPCR of Polymers adenine tail method.(2) The relative expression level of miR-21is1.101×-3±0.487×10-3in BPE and2.283×10-3±0.898×10-3in NSCLC with MPE.The expression level of miR-21was significantly higer in NSCLC with MPE than benign pleural effusion (P<0.05). The relative expression level of miR-126is0.687×10-3±0.172×10-3in BPE and0.466×10-3±0.162×10-3in NSCLC with MPE.The expression level of miR-126was significantly lower in NSCLC with MPE than benign pleural effusion (P<0.05)(3) The AUC for expression level of miR-21in pleural effusion was0.837,95%CI0.724to0.950. The AUC for expression level of miR-126in pleural effusion was0.810,95%CI0.693to0.927.Conclusion:(1) The expression of miRNAs in pleural effusion could be tested by qRT-PCR.(2) The expression level of miR-21was showed higher levels in pleural effusion from NSCLC patients with malignant, but the level of miR-126was decreased.(3) MiR-21and miR-126in pleural effusion can be used as potential biomarkers of diagnosis of NSCLC patients with malignant pleural effusion.
