Objective: this study through the detection of VEGF, MMP- 9 and CD44 in non-small cell lung cancer with expression level of malignant pleural effusion and the mi RNA expression, preliminary understanding from the molecular level VEGF, MMP- 9, CD44 and micrornas with lung cancer with malignant pleural effusion, the relationship between the formation mechanism of malignant pleural effusion provide some possible laboratory basis.Methods:1. Using the technology of enzyme-linked immunosorbent method respectively detect VEGF, MMP- 9 and CD44 in 25 cases of non-small cell lung cancer with malignant pleural effusion and non cancerous pleural effusion expression of 16 cases of two groups of specimens, analyze its expression level with NSCLC different lymph node staging, pathological classification, and three indicators of expression in lung cancer with MPE whether has some contact.2.Application of gene chip technology, malignant pleural effusion and benign pleural effusion was detected in the gene expression spectrum of 1 case specimens using microarray analysis screen out the significant difference of micrornas, of the target gene by fluorescence quantitative PCR method for further verification.Results:1) The VEGF, MMP- 9 and CD44 expression in malignant pleural effusion and benign pleural effusion were statistically significant(p < 0.05);In malignant pleural effusion, VEGF, MMP- 9, and there is a positive correlation between CD44, and three indicators in the benign pleural effusion, no correlation.In different pathological classification, VEGF, MMP- 9, and the expression of CD44 in lung adenocarcinoma, levels higher than the lung squamous carcinoma(p < 0.005).Different lymph node staging, VEGF, MMP- 9, and the expression of CD44 in N3 stage N2 were increased to a certain extent, but there was no significant difference statistically(p > 0.05).2) Benign and malignant pleural effusion gene expression spectrum of micrornas microarray analysis, expression differences in 2.0 there are 312 times more micrornas, the expression level of 216, 96 cut.To show significant difference of micrornas- 423, the mi RNA- 486 using the fluorescent quantitative PCR method for validation, the purpose gene fusion curve for single piece, better amplification specificity, mi R- 423 relative expression in malignant pleural effusion in quantity, the expression of mi R-486 is higher.Conclusion:1. VEGF, MMP- 9, and increased expression of CD44 in lung cancer with malignant pleural effusion.Closely associated with the formation of MPE;Have a certain relevance and pathological type, lymph node staging.Three indicators of joint detection identification of benign and malignant pleural effusion and pathological classification, early judgment has a certain reference value.2.
