Objective To compare the value of18F-FDG,"C-MET and "C-CHO PET/CT for differentiating gliomas from acute or chronic inflammation in experiment rat models. And to investigate the relationship of18F-FDG,11C-MET,11C-CHO uptake with GLUT-1, HIF-la, VEGF, Ki67and MMP-9in C6glioma.Methods Part Ⅰ:①36male Wistar rats were randomly divided into5groups:group A or B consisted of8rats bearing both C6glioma and turpentine oil-induce acute inflammation; group C or D consisted of8rats bearing both C6glioma and turpentine oil-induce chronic inflammation; group E as a control group consisted4blank rats.②11C-MET and18F-FDG PET/CT were performed on group A and C;11C-CHO and18F-FDG PET/CT were performed on group B and D;3tracers of PET/CT were performed on group E. The lesion-to-muscle(L/M) ratios and the tumor selectivity index were calculated.③A one-way ANOVA, a unpaired t test were used to make statistical analyses. Part II:①fter the examinations, the lesions were excised. Immunohistochemical stain was used to demonstrate the situation of GLUT-1, HIF-1α, VEGF, Ki67and MMP-9.②A multiple regression and correlationwere used to make statistical analyses.Result①The18F-FDG uptake of C6glioma, acute inflammation and chronic inflammation were higher than the uptake of11C-MET and11C-CHO, and the differences were significant. The uptake of11C-MET and11C-CHO in all the lesions had no significant difference.②The18F-FDG and "C-MET uptake of C6glioma were higher than acute and chonic inflammation, and the differences were significant. And the SIMET is significantly higher than SIFDG The FDG and MET uptake of the two inflammations had no significant difference.③The CHO uptake from high to low level in different lesions is:C6glioma>acute inflammation>chonic inflammation.And there were no significant difference between different lesions.④The Tu/Mu-FDG correlated with GLUT-1, HIF-1α, VEGF, Ki67, MMP-9in order.⑤The Tu/Mu-MET correlated with VEGF, Ki67, MMP-9, HIF-1α in order. There were no correlations between Tu/Mu-MET with GLUT-1.⑥The Tu/Mu-CHO correlated with Ki67, VEGF in order. There were no correlations between Tu/Mu-CHO with GLUT-1, MMP-9and HIF-1α.Conclusion18F-FDG and "C-MET PET/CT are able to differentiate C6glioma from inflammations, and11C-MET PET/CT is more tumor selective, therefor it is suitable for clinic to diagnose glioma.11C-CHO PET/CT is not able to differentiate glioma from inflammations, therefor its clinical value is limited. The18F-FDG,11C-MET,11C-CHO uptake of C6glioma correlate with tumor biologic character, such as glucose transform, hypoxia, proliferation, angiogenesis and invasiveness.The18F-FDG uptake is influenced by glucose transform, hypoxia, angiogenesis, proliferation, invasiveness, in order. The "C-MET uptake is influenced by angiogenesis, proliferation, invasiveness, hypoxia, in order. The11C-CHO uptake is influenced by proliferation, angiogenesis, in order.
