Objective:To explore the significance of HER-2protein, CyclinDl protein in papillary thyroid carcinoma (PTC). To investigate the correlation between HER-2protein overexpression and HER-2gene amplification in papillary thyroid carcinoma.Methods:Immunohistochemical staining was performed in97specimens of PTC and97specimens of adjacent benign lesions to examine the expression of HER-2protein and CyclinDl protein. FISH was employed to detect the HER-2gene amplification of18samples with HER-2of2+/3+in PTC.Result:1. The positive ratio of HER-2in PTC was46.2%, and no expression in the adjacent benign lesions. There were all statistical difference between them(p<0.05). HER-2expression correlation with clinicopathological parameters analysis:HER-2expression correlated with tumor size only in PTC, HER-2was not expressed in less than or equal to lcm in diameter of thyroid papillary carcinoma. There were no relationships between expression of the maker and other clinicopothological features in PTC.2. There were4positive cases and14negative cases detectecd in18samples of PTC by fiuorescence in situ hybridization. Compliance rate was showed22.2%between HER-2gene amplification and HER-2protein expression.3. The positive ratio of CyclinD1in PTC was92.8%, and no expression in the adjacent benign lesions. There were all statistical difference between them(p<0.05). CyclinD1was seen in92.8percentage of papillary thyroid carcinoma, the specificity can reach100%. There were no relationships between expression of the maker and the clinicopothological features in PTC. Preliminary observations indicate that heterogeneity of CyclinD1expression in cancer tissue and may be associated with lymph node metastasis.4. There were no relationships between expression of HER-2and CyclinDl in PTC.Conclusion:1. HER-2protein overexpression and HER-2gene amplification play an important role in the carcinogenesis of PTC. Her-2could be one of potential targets for targeted therapy in PTC.2. The correlation between them HER-2protein overexpression and HER-2gene amplification show positive correlation in PTC, which as a potential target for targeted therapy and its diagnostic criteria still need to be further explored.3. CyclinD1could be as a molecular marker of early diagnosis in papillary thyroid carcinoma. The relationships of CyclinDl’s heterogeneous expression and lymph node metastasis are worth to further exploration.
