Objective : To investigate the expression of CyclinD1 and anti-oncogene PTEN in papillary thyroid carcinoma(PTC) and its clinicopathologic significance.And further analyze the expression trends of this two protein and the correlation between the BRAFV600 E gene mutation,preliminary reveal the role of this two protein in the carcinogenesis of PTC.Methods:EnVision two step immunohistochemical method was applied to detect the expression levels of CyclinD1 and PTEN in the 70 cases of PTC tissues.PCR and DNA sequencing was used to detect the BRAFV600 E gene mutation in these PTC tissues,then analyze the correlation of the BRAFV600 E gene mutation and the clinicopathological features of PTC with the expression of CyclinD1 and PTEN protein.Results:In this 70 cases,the positive CyclinD1 expression rate in thyroid papillary carcinoma tissue was 77.1%(54/70) and in tumor-adjacent normal tissue was 12.8%(9/70), the difference express was statistically significant(x~2 =71.957,P<0.00);The up-regulation of CyclinD1 was associated with tumor lymph node metastasis and pathological stage,but has no correlation with BRAFV600 E gene mutation(r=0.625,P>0.419),age,sex and tumor diameter.The negative PTEN protein expression rate in thyroid papillary carcinoma tissue was 78.6%(55/70) and in tumor-adjacent normal tissue was18.6%(13/70), the difference express was statistically significant(x~2 =50.44,P<0.00);The down-regulation of CyclinD1 was associated with sex,lymph node metastasis and pathological stage,but has no correlation with BRAFV600 E gene mutation(r=1.349,P>0.238),age and tumor diameter.In this 70 cases of PTC,the BRAFV600 E gene mutation rate is 60% and was closely related to lymph node metastasis,pathological stage(x~2 =37.333;P<0.05),(x~2 =35.700;P<0.05),but has no correlation with age,sex and tumor diameter.Conclusion:The expression of CyclinD1 and PTEN protein has no linear relationship.They may be an independent risk factor for poor prognosis of PTC, and maybe used as a marker for evaluating the prognosis of PTC.They may guide the individual clinical treatment and prognosis of PTC.
