Expression Of MDM2and PTEN In Gastric Biopsy Of Cardia Carcinoma

Background Cardia carcinoma is the most common gastrointestinal tumor,invasion, metastasis and recurrence is an important factor to affect the prognosis. Inthis process, many oncogenes, adhesion molecules involved in tumor-associatedproteases and their inhibitors. Cardia mechanism is unclear, many scholars study itfrom different angles. Tumor development process, the activation of oncogenes,tumor suppressor gene abnormalities.Murine double minute2(MDM2) gene is aproto-oncogene, MDM2contain only small amounts in normal tissue, undetectable byimmunohisto-chemical methods.Study found that MDM2gene mutation,amplification and abnormal expression in a variety of tumor tissues. Human tumorsin36%of sarcoma and10%of well differentiated glioma, as well as a small part ofesophageal cancer, neuroblastoma, the degenerative astrocytoma MDM2geneamplification. PTEN is a dualspecificity lipid phosphatase Originally identified asatumor suppressor genes found so far. PTEN mainly through its dualsubstratespecificity phosphatase activity, via the PI3K signaling and MAPKsignalcascades play a tumor inhibition of cell growth, apoptosiproliferation and adhesion ofan impact. In this study,using immunohisto-chemical method detection of MDM2, theexpression of PTEN in the gastric cardia, and to further explore the relationshipbetween the development of cardia cancer.Aim Study on relationship between MDM2，PTEN gene and progression ofcardiac carcinoma.Methods Cardiac cancertissues（n=32）, nomal cardiac tissues（n=18）weredetected for MDM2and PTEN expression by S-P immunohistochemistry methods. Results The MDM2protein expression in cardiac carcinoma tissues was higherthan the normalcardiac tissues （P<0.05），and was related statistically withlymphote metastasis and depth of invasion（P<0.05）. The PTEN protein expressionin cardiaccarcinoma tissues was lower than the normal gastric tissues （P<0.05），and was related statistically with lymphotemetastasis and degree of differentiation（P<0.05）.Conclusion Abnormal expression of MDM2and PTEN is closely associated withcardiac carcinoma, which may be contribute in the tumorigenesis, growth andmetastasis of gastric carcinoma.
