Gallbladder cancer is one of the most common malignancies in biliary system,Inrecent years, its incidence is rising. Most of the early symptoms of gallbladder cancerpatients due to the lack of specificity are easily overlooked, patients have beendeveloped to advanced symptoms at time of diagnosis, operation is difficult to radicalresection at this time,5years of survival rate is low. For the time being, theoccurrence and development of mechanism has not been completely clear. Autophagyas Ⅱ type of programmed cell death, experimental studies have been confirmed thatthe occurrence, progression, tumor angiogenesis in tumors and autophagy have closerelationship. Therefore, we research autophagy related gene in the occurrence anddevelopment process of gallbladder cancer, explore the mechanism and correspondingtreatment, have important clinical significance and research value.Objective: By detecting gallbladder carcinoma(GBC)tissues autophagy relatedgenes beclin1and microtubule-associated protein1light chain3-B (MAP1LC3B)expression, with clinical and pathological parameters, observe autophagy relatedgenes beclin1and MAP1LC3B mutual relations in gallbladder carcinoma and try tofind the molecular markers about gallbladder cancer, at the level of molecular biologyto provide scientific basis for early diagnostic and effective treatment of gallbladdercarcinoma.Methods:forty-six cases of gallbladder carcinoma wax in the First AffiliatedHospital of Shanxi Medical University in January2008to December2011werecollected,46cases of cholecystitis over the same period were matched groups, expression of various groups of Beclin1and MAP1LC3B protein were detected byimmunohistochemical method in gallbladder cancer and cholecystitis and analyze therelationship between the clinical and pathological features. Data is analysed bychi-square test and Spearman rank correlation analysis of SPSS17.0software.Result:1. The positive rate of Beclin1was45.7%in gallbladder carcinoma wassignificantly lower than the expression in control group, which was78.3%(P <0.05).The positive rate of MAP1LC3B was47.8%in gallbladder carcinoma wassignificantly lower than the expression in control group, which was71.7%(P <0.05).2. The expression of Beclin1and MAP1LC3B in gallbladder carcinoma wascorrelated with lymph node metastasis and clinical stage (P<0.05). The expressionof Beclin1and MAP1LC3B was not correlated with gender, age, degree ofdifferentiation (P<0.05).3. There was a significantly positive correlation was discovered between theexpression of Beclin1,MAP1LC3B in gallbladder carcinoma (P <0.05).Conclusion:1. The positive rates of Beclin1was45.7%in gallbladder carcinoma wassignificantly lower than the expression in control group. The positive rates ofMAP1LC3B was47.8%in gallbladder carcinoma was significantly lower than theexpression in control group.2. There was a significantly positive correlation was discovered between theexpression of Beclin1,MAP1LC3B in gallbladder carcinoma (P <0.05).3. The expression of Beclin1and MAP1LC3B in gallbladder carcinoma wasconnected with clinical stage,lymph node and far diversion(P<0.05).4. The expression of Beclin1, MAP1LC3B were independent factors in theprognosis of gallbladder carcinoma patients,and There was a positive correlation between Beclin1and MAP1LC3B expression. Combined detect the expression ofBeclin1, MAP1LC3B might be an important reference index for the earlydiagnosis,analysis prognosis of gallbladder carcinoma.
