Introduction:Anterior thalamic nucleus is an important part of the dorsal thalamus,which is composed of three sub-nucleuses, including dorsal nucleus anterior thalamicnucleus (AD), anterior thalamic nucleus ventral nucleus (AV) and intermediate nucleusanterior thalamic nucleus (AM). Its function is to provide relationship between spatiallearning and memory. P-CREB is a der transkriptionsfaktor which plays an importantrole in the process of spatial learning and memory.Objective: In this study we use Morris water maze behavioral experiment andimmunohistochemistry (IHC) of P-CREB, to observe the expression and distribution ofP-CREB in rat anterior thalamic nucleus, as P-CREB participate in learning andmemory process, provide morphological data to anterior thalamic nucleus.Methods: Animals used in this experiment are thirty Wister rats, male, weighed200-250g. These rats were tended for five days in the Morris water maze behavioralexperiments, After6days anesthetized by intraperitoneal injection of diethyl ether and4%chloral hydrate, with scissor expose chest cavity of rat, infusion needle inserted inthe left ventricle of Heart into the ascending aorta perfuse0.9%NaCl and4%Paraformaldehyde. Brain perfusion is completed immediately, placed in4%paraformaldehyde fixative overnight at4°C. The next day using the continuousvibration slicer coronal slices, slice with thickness of30μm. Using the anti-CREBantibody as a marker, employ SABC immunohistochemical staining, observe theexpression and distribution of P-CREB, help out in learning and memory process, inanterior thalamic nucleus. All specimens were photographed and analyzed by same setof image analysis system, the resulting data assessed by SPSS17.0software paired t-test. Results:1. Morris water maze behavioral test results: After four days of spacenavigation test, experimental group pass through Escape Latency test,2,3,4days“Escape latency” compare with the first day Escape latency, has notable shorten time(p<0.05), explanation of experimental rats with a certain spatial learning and memory,with increase in the number of spatial positioning navigation training, spatial learningability of rats were increased, by learning and spatial orientation successfullyestablished a spatial orientation memory.2. P-CREB immunohistochemical results: Through light microscope, observed theexpression of p-CREB located within the nucleus, circular and oval, and noticeableexpression in both AV and AD zone. Experimental group P-CREB immunoreactiveproducts average optical density values in AD and AV area compared with control grouphas statistically significant difference(p<0.05). P-CREB expression in AD zone comparewithAV zone in experimental group, was not significantly different (P>0.05).Conclusion: In the process of learning and memory, P-creb expressed in ratanterior thalamic nucleus.
