Backgrounds and Objectives:Currently, the high incidence of lung cancer in the world, has created a significanthuman health threat. With the acceleration of modernization and urbanization process inChina, lung cancer is our country’s highest incidence of malignant tumors, its incidence,mortality, the highest mortality rate of men and women are in the first place. Lungcancer is a malignant tumor of the higher, which occurrence is the process ofmulti-stage development that more than one factor are involved in, the incidence of lungcancer is often very hidden, therefore, many patients are already in an advanced stage.So, early diagnosis, intervention and treatment of lung cancer is very important.Metastasis-associated protein S100A4(S100calcium binding protein A4), is animportant member of the S100protein family, in recent years, it has received muchattention in the research area. Studies have shown that, S100A4involves in the processof abnormal proliferation,malignant transformation, invasion, angiogenesis of cells,by increasing athletic ability of tumor cells, reducing the adhesion between tumor cells,promoting the degradation and remodeling of extracellular matrix, promoting tumorangiogenesis, inhibiting apoptosis,and promoting abnormal cell proliferation. Its role inpromoting tumor with invasion of a variety of malignant tumors, metastasis shows apositive correlation. Currently, S100A4in the tissues has been confirmed to monitor the biological behavior of cancer as an independent factor. However, the study that S100A4protein levels in human peripheral blood associated with the development of cancer isfew.This paper aims to determine lung adenocarcinoma, lung squamous cell carcinomaof S100A4levels in the peripheral blood, by using double antibody sandwichenzyme-linked immunosorbent assay (ELISA), to study its correlation with lungadenocarcinoma and lung squamous cell carcinoma. Thus, this research may providemore valuable data, by understanding S100A4expression in peripheral blood of patientswith tumors, in order to determine whether it can become a new type of lung cancerserum tumor markers.Materials and Methods:Blood samples collected in this study, come from158patients in th the AffiliatedHospital of Dalian Medical University from November2013to April2014.Including104cases of with lung cancer in the thoracic surgery ward, of which89cases of lung adenocarcinoma,15cases of lung squamous cell carcinoma,and21cases of lung cancer patients receiving chemotherapy in the oncology ward,33cases ofmedical patients.For lung cancer patients in the thoracic surgery, We extract blood before surgery,and save their peripheral blood after centrifugation, Subsequently, blood samples areselected and grouped according to the results of the pathological findings. Ultimately,we measure the S100A4level in the samples by ELISA.Analyse the resulting data usingSPSS17package.The final result reveal the relationship between S100A4levels in peripheral bloodand lung cancer, as well as in different pathological types of lung cancer, such as lungadenocarcinoma and lung squamous cell carcinoma of the level difference.Results:1.In the sampling period, the number of cases of lung squamous cell carcinomawas significantly lower than the lung adenocarcinoma. The incidence of lungadenocarcinoma is higher in women than in men, lung squamous cell carcinoma is higher in men than in women, and they are statistically significant (P <0.05).2.In the medical group,S100A4protein in the peripheral blood is55.82±16.68ng/ml, in the lung adenocarcinoma, it is85.19±22.46ng/ml.In lungsquamous cell carcinoma, it is53.8±20.35ng/ml. S100A4protein levels in lungadenocarcinoma was significantly higher than normal physical examination, with astatistically significant difference (p <0.05) between the two. However, there is nodifference in lung squamous cell carcinoma than in the normal group.3.In the lung squamous cell carcinoma group, when there is more poorly extent,larger tumor size, S100A4protein levels are significantly higher, and it is statisticallysignificant (P <0.05). There are differences in terms of pathologic stage, but was notstatistically significant (P>0.05).4.In the lung adenocarcinoma group, when there is higher pathological stage, widerinfiltration case, S100A4protein levels are significantly higher, and it is statisticallysignificant (P <0.05). There are differences in the degree of differentiation and tumorsize, but not statistically significant (P>0.05).Conclusion:1.Compared with the medical group, S100A4protein levels in the peripheral bloodof patients with lung adenocarcinoma group are significantly increased, while lungsquamous cell carcinoma group did not change significantly.2.S100A4protein levels in peripheral blood,which is in Lung adenocarcinoma andlung squamous cell carcinoma group, are significantly different, and S100A4proteinlevels show a correlation with pathological staging, degree of differentiation, tumor size,depth of invasion.3.S100A4can be used as a novel serum tumor marker for the detection of lungadenocarcinoma.
