ObjectiveTo study the expression of RhoA/RhoB and Rock1/Rock2in placentatissue of severe preeclampsia (sPE) in the hope of making the mechanism of sPEmore clear.Methods30placenta specimens of sPE were collected from obstetrical department ofTengzhou central people’s hospital as the experimental group; the controlspecimens were placenta tissue of normal-term cesarean delivery. The diagnosisstandard of sPE patients refers to the seventh edition of obstetrics andgynecology. The location and expression of RhoA/RhoB and Rock1/Rock2between the two groups were detected by Immunohistochemistry.Results1. RhoA were detected in cytoplasm of the trophoblasts, endothelial cellsand endometrial stromal cells both in the two groups, mainly in cytoplasm of thesyncytiotrophoblast, similar to the location of RhoA. And the level of RhoA insPE group was up-regulated contrast to the control group(p<0.05)2. RhoB were detected in cytoplasm of the trophoblasts, endothelial cellsand endometrial stromal cells both in the two groups, mainly in cytoplasm of thesyncytiotrophoblast, similar to the location of RhoB. And the level of RhoB insPE group was up-regulated contrast to the control group(p<0.05)3. Rock1mainly located in the cytoplasm of syncytiotrophoblasts, was alsodetected in cytoplasm of cytotrophoblast, and less expressed in nucleus,endothelial cells and endometrial stromal cells. The level of Rock1weresignificantly higher in the sPE group than that in control group.4. Rock2mainly located in the cytoplasm of syncytiotrophoblasts, was also detected in cytoplasm of cytotrophoblast, and less expressed in nucleus,endothelial cells and endometrial stromal cells. The level of Rock2weresignificantly higher in the sPE group than that in control group.ConclusionThe signal path consists of upstream molecular RhoA/RhoB anddownstream molecular Rock1/Rock2appeared up-regulated in sPE, whichdemonstrates the signal moleculars of RhoA/RhoB/Rock1/Rock2may involvein the pathogenesis of sPE through affecting shallow placenta implantation inpreeclampsia, ischemia, hypoxia and apoptosis of trophoblasts, injury ofvascular endothelial cells and artery vasospasm.
