Objective:To approach the treatment effect of prostate cancer PC-3cell lines with combination of All-Trans-Retinoic Acid (ATRA) and Docetaxel (DOC).Methods:The cells could be divided into three groups, include negative group, ATRA group and DOC group. The growth inhibition ratios were measured by MTS assay, The cells cycle and apoptosis rates were examined by flow cytometry (FCM), The cell morphological changes were examined by ordinary optical microscopy, The CD133+CD44+cells were examined by flow cytometry (FCM).Results:The ATRA or DOC could effectively inhibit the proliferation of the PC-3cells as a concentration-dependent manner, the inhibitory rate of combination treatment with ATRA and DOC was significantly higher than that of ATRA or DOC repectively,(P <0.05); Typical morphological changes could be observed by using ordinary microscope after treatment with ATRA and DOC, including nuclear chromatin condensation, fragmentation, cell shrinkage and formation of apoptosis bodies, combination treatment group is more obvious. It was further confirmed by flow cytometry that the inductive effect on cell apoptosis became more obvious than that ATRA or DOC alone, when ATRA combined with DOC (P<0.05). It was confirmed by flow cytometry that CD133+CD44+cells was reduce by used ATRA (P<0.05).Conclusion:ATRA combined with DOC have obvious synergistic effects in inhibiting the proliferation of PC-3cells and enhancing the effects on inducing apoptosis. The stem cells of PC-3line might be increased by use DOC. The stem cells of PC-3line might be induced by used ATRA.
