ObjectiveSpinal cord injury (SCI) is the most serious injury of the central nervous system, regeneration mechanism of nerve and glial after injury has not yet been understood. Neural stem cells (NSC) transplantation is an effective strategy for the treatment of SCI, but the exogenous NSC transplantation have problems of tumorigenicity, different differentiation, tissue compatibility. The effect of electric-acupuncture in the treatment of spinal cord injury has been affirmed. Notch signaling pathway plays a permissive role in the proliferation, differentiation process of neural stem cell (NSC). In this study, we use an aneurysm clip to make spinal cord injury (SCI) model of SD rats.to research the expression of Notchl protein in spinal cord injury SD rats after the treatment of electric-acupuncture. to detect whether electric-acupuncture have an effect on spinal cord injuries through the Notch signaling pathway.Methods1.120SD rats were randomly divided into spinal cord injury group, electro-acupuncture group and the sham group, Clamping T10spinal cord segmental in the injury group; Control group animals opened the vertebral canal, electro-acupuncture group. Electro-acupuncture group began taking treatment on3days after surgery. Record BBB behavior score respectively at Id,3d,5d,7d,14d,21d,28d after surgery, assess neurological recovery.2. Draw the materials of each group at7d,14d,21d,28d after surgery. with immunohistochemistry and Western blot analysis, research the expression of notch-1protein and proliferation of endogenous stem cell after spinal cord injury.3.Using SPSS Statistics17.0statistical software for analysis, and the data of each group using paired samples t test to α=0.05as a test. Result1. BBB score data suggest that, data of control group is significantly higher than injury group; Each set of data increased gradually with the growth of the postoperative time; Different time points within the same group had significant difference (P<0.05). At the same time points of comparison between different groups had significant difference (P<0.05).2. Between different groups at the same time point, the number of endogenous neural stem cells compared to the EA group was significantly higher than other groups; contrast between each group were significantly different (P<0.05). Comparison between different time points in EA group, the number of cells growing with time; comparison between different time points were significantly different (P<0.05).3. Between different groups at the same time point, the expression of Notchl protein in EA group was significantly lower than the injury group; contrast between EA, injury group and the sham group were significatly different (P<0.05). Comparison between different time points of EA group, the expression of Notchl protein decreased with the time grew; comparison between different time points were significantly different (P<0.05).Conclusion1. Electro-acupuncture can significantly improve BBB behavior score, from the perspective of behavior can be interpreted as electro-acupuncture treatment of spinal cord injury has obvious therapeutic effect.2. Electro-acupuncture can induce proliferation of neural stem cell after spinal cord injury in the rat.3. Electro-acupuncture can inhibit the expression of Notchl protein after spinal cord injury.4. Electro-acupuncture may inhibit the expression of Notchl after spinal cord injury while eliminating molecules signal of neural stem cells into glial cells.
