Objective The development of Nasopharyngeal Carcinoma was associated with Epstein-Barr virus infection. Epstein-Barr virus infection can lead to the change of chromogranin A protein expression level in serum. This study was perform to investigate the diagnostic value of measurement of serum Epstein-Barr virus DNA(EBV-DNA)、 serum sialic acid SA(SA)、chromogranin A(CgA) and Epstein-Barr virus-related antibodies. Epstein-Barr virus-related antibodies include Epstein-Barr virus early antigen immunoglobulin A (EBV-EA-IgA)、Epstein-Barr virus capsid antigen immunoglobulin A (EBV-CA-IgA) and Epstein-Barr virus cleavage product of the immediate early gene expression BRLF1of Rta antibody IgG (Rta-IgG). Methods Serum EBV-EA-IgA、 EBV-CA-IgA、Rta-IgG、EBV-DNA、SA and CgA levels were measured in249specimens of patients with Nasopharyngeal Carcinoma,50patients with EBV-CA-IgA positive,50patients with rhinitis and50controls, respectively. Results Six markers levels in patients with nasopharyngeal carcinoma were significantly higher than the control groups. Single positive marker as a diagnostic criterion, the sensitivity of EBV-EA-IgA、 EBV-CA-IgA、Rta-IgG、EBV-DNA、SA and CgA was31.37%’76.47%、67.65%.64.71%、58.82%and74.51%respectively. The specificity was96.0%.94.0%、98.0%、98.0%,84.0%and54.0%respectively. With combined detection of two or more markers, the sensitivity would be96.08%and the specificity would be95%. Conclusion Measurement of serum EBV-EA-IgA、EBV-CA-IgA、Rta-IgG、EBV-DNA、SA and CgA levels is of clinical usefulness for diagnosis of Nasopharyngeal Carcinoma. The marker EBV-DNA can reflect the dynamic development of nasopharyngeal carcinoma. It can be used to monitor the efficacy of treatment of patients with nasopharyngeal carcinoma.
