Objective:To investigate the role of LKB1/AMPK/mTOR signal transductive pathway in the occurrence of endometrial cancer, in order to elucidate the pathogenesis of experimental basis of endometrial carcinoma.Method:In30cases of normal endometrium as control group,endometrial carcinoma group, endometrial carcinoma,atypical hyperplasia of endometrium30cases as experimental group. LKB1,AMPK, mTOR expression was detected in the tissue with immunohistochemistry method.Results:1.Three groups of patients with LKB1, AMPK, mTOR expression:in normal endometrium, atypical hyperplasia of endometrium, endometrial cancer, the positive expression of LKB1, AMPK decreased gradually, the expression of LKB1, AMPK in endometrial carcinoma group were lower than those of atypical hyperplasia group and proliferative endometrium group (χ2=5.711, P<0.05;χ2=19.288, P<0.01; χ2=4.593, P<0.05;χ2=17.067, P<0.01), while the expression of atypical hyperplasia group LKB1, AMPK were lower in proliferative endometrium group (χ2=4.8, P <0.05;χ2=4.39, P<0.05). The positive expression rates of mTOR were increased, mTOR expression in endometrial carcinoma group was higher than that of atypical hyperplasia and proliferative endometrium group (χ2=5.079, P<0.05;χ2=26.667, P <0.01), the expression of mTOR was higher than that of atypical hyperplasia of endometrium group (χ2=10.33, P<0.01).2.Three groups of patients with LKB1, AMPK,mTOR expression:in proliferative with LKB1, AMPK, mTOR expression:in proliferative endometrium, endometrial atypical hyperplasia and endometrial adenocarcinoma,LKB1positively correlated with AMPK expression (r=0.999, P<0.01),LKBI, AMPK negatively correlated with mTOR expression(r=-0.989, P<0.01; r=-0.989, P<0.01).3.The correlation between LKB1, AMPK, mTOR expression and clinical pathological characteristics of endometrial carcinoma:LKB1, AMPK expression in endometrial adenocarcinoma and was related to the histological grade, regardless of patient age, FIGO staging and operation pathology, the mTOR expression in endometrial adenocarcinoma and histological grade and the vast by state, regardless of stage and the age of patients and operation pathology.Conclusion:The results of this study confirmed that, LKB1/AMPK/mTOR signal transduction pathway plays an important role in the occurrence and development of endometrial cancer, it may become a new target for treatment of endometrial cancer.
