Objective:Hepatic ischemia reperfusion injury is a complex, multi factor process, including inflammation, oxidative stress and apoptosis which were harmful to the liver cells. Berberine is a ueaful drug for anti-inflammatory. Recent studies showed that berberine also has the anti oxidative stress function and inhibit cell apoptosis function. The purpose of this experiment was to study the protective effect of berberine on hepatic ischemia reperfusion injury in rats and the mechanism.Methods:24healthy male SD rats, age8-10weeks, body weight200250g, were randomLy divided into3roups (n=8):Sham operation group (group A), control group (group B) and berberine group (group C). Group A only needs laparotomy operation.14days before operation Physiological saline is given to rats by gastric gavage in group B.14days before operation berberine is given to rats by gastric gavage in group C. B, C group were used a modified Hu Jianping method for in cold ischemia reperfusion. Hu Jianping method was modified in the experimental, changing the inflow tract splenic vein to the portal vein and the outflow tract of right adrenal vein to inferior vena. The outflow tract diameter was increased, avoid the occurrence of liver reperfusion edema.6hours after the operation, blood samples is collected for testing the content of ALT and AST in rat, using ELISAto measure with IL-1β, IL-6of liver inflammation; liver tissue, to observe the morphological changes of liver tissue; MDA and SOD were tested in ELISA method, to evaluate the liver tissue damage which is induced by oxidative stress injury; Detecting of liver tissue Caspase-3activity and, TUNEL method for detection of apoptosis. The data was used SPSS17.0for statistical analysis. Results: the serum of ALT, AST, IL-1β, IL-6and content of MDA in Group A were lower than that in group B, C (P<0.01), the content of SOD in the liver tissue of group A was higher than that in group B, C (P<0.01). The serum of ALT, AST and IL-1β concentrations in groups B and C were lower (P<0.05). Compared with group B, the content of MDA in liver tissue were decreased (P<0.05) and SOD in liver tissue was increased in group C (P<0.05). Expression of apoptosis and capase-3in group C were lower than group B (P<0.05). The group A was the normal structure of liver tissue, the liver tissue is showed varying degrees of damage in groups B and C. There was obvious injury of liver tissue in group B, the injury of liver tissue was relatively light in group C.Conclusion:Rats liver cold ischemia reperfusion model was used in this study including cold perfusion, reflow process. This study can imitate the donor liver perfusion of liver transplantation, replantation process.6h after reperfusion transaminase was lifted. Injury and hepatic histopathology suggested that model was successful. Berberine has a protective effect on liver cold ischemia reperfusion injury in rats, the mechanism is that berberine can be restrain the liver inflammation and oxidative stress reaction, and reduce the apoptosis of liver cells.
