Objective:To investigate the protective effect of KGF and G-CSF on mice irradiated by5.0Gy gamma ray and then do some research on the mechanism. The purpose of our research is to provide an important theoretical basis for cytokine combination therapy on radiation injury.Methods:C57BL/6mice were selected to receive whole body irradiation of37Cs-γ ray to build a mouse model of acute radiation sickness. Test the effects on the blood and immune system in irradiated mice by the indexes of body weight, organ indexes, white blood cells, red blood cells, concentration of hemoglobin and blood pelets in peripheral blood, colony forming units of spleen and so on. Test the effects on bone marrow hematopoietic system in irradiated mice by the indexes of bone marrow nucleated cells, bone marrow DNA contents, listeria-granulocyte colony forming unit (CFU-GM), red blood cell blowout type colony generating unit (BFU-E) and so on. Detect the activity of superoxide dismutase (SOD) and content of Maleic Dialdehyde (MDA) to reflect its effect on antioxidation in irradiated mice.Results:The weight of combined drug group is significantly recovers better than other groups. Compared with simple control group, combined drug group can reduce the damage to the peripheral blood caused by radiation. Joint drug delivery can improve thymus coefficient, spleen coefficient, the number of CFU-S irradiation mice, and protect the immune system in radiated mice. Joint drug delivery can ameliorate BMNC, bone marrow DNA content, CFU-GM, BFU-E caused by radiation obviously, and protect the bone marrow hematopoietic system from radiation damage. Joint drug delivery can increase the SOD activity in liver tissue, decrease the MDA content, and protect the liver tissue caused from radiation damage.Conclusions:Different drug groups are of different protective effects on hematopoietic system, immunity system and the organization organs of irradiation mice. This study indicates that KGF in combination with G-CSF is of some protective effect from radiation damage on C57BL/6mice.
