ObjectTo explore the effect of the expanded capsule on the grafts of autogenous costal cartilage, including the general condition, the histomorphology, the ultrastructure. We design this experiment in order to provide the theoretical basis for the clinical surgery about the expanded capsule.MethodsSixteen New Zealand white rabbits aged3months were involved and four15ml tissue expanders were implanted on the back symmetrically each rabbit. After the surgery, at the time of the7thday, we inject the normal saline in the expander, total30ml. After1month, the eapanded capsule formed, the tissue expanders were removed, the left sides removed the capsule (experimental group), the right sides reserved(controlgroup) meanwhile the6th and7th costal cartilage without the perichondrium were taken out, devided into two pares and weighed each, and then placed on the two groups symmetrically. Four weeks and8weeks later, obtained the cartilage, weighed all the grafts, observed the general condition, histomorphologic and ultrastructure examination.ResultsThe cartilage showed more obvious increase in weight in the experimental group than the control group, and there are statistical significance between the4th week and the8th week of both the experimental and the control group, the weight change are (0.0034±0.0027)、(0.0058±0.0014) g,(-0.0003±0.0019)、(-.0039+0.0053) g.Microscopic observation:the number of the cartilage cell (HE400X) in the experimental and control group at the4th and8th week after the transplantation increased, which are (62.0±3.2)、(68.2±3.5),(51.8±1.4)、(58.4±7.1), there are statistical significance. And the colour are deeper in the experimental than the control group. Electron observation:In the experimental group, as the time going on, the cytomembrane maintained intact, the cell volume growed, and the rough endoplasmic reticulum and the mitochondria decreased, the nucleus begin edged, the lipid droplet growed and mixed together, and appeared impurity at the8th week after the transplant-ation. In the control group, the same change happened, but the changes happened distinct and rapid.Conclusion:Removed the expanded capsule during the operation is benefical to the the grafts of autogenous costal cartilage, in which the weight of the grafts and the number of the cartilage cell increased more significiently. The costal cartilage can survive and develop after transplantion without the perichondrium. The results can provide corresponding experimental guidance for the clinic problems.
