Purpose and significanceAllergic rhinitis and nasal polyps are common allergic reactions and related diseases, with the incidence in the normal population generally higher. Relations of pathogenesis between the two diseases have long been the focus of basic and clinical research, and so far still not fully clarified. In recent years, more and more evidence suggests that the incidence of allergic rhinitis involves regulations of multiple genes, and the pathogenesis of nasal polyps involves the expression change of multiple genes. In this research, through histological test and exploring the gene expression profiles of allergic rhinitis and nasal polyps by RNAseq technology, the differences of pathogenesis and gene expression profiles between allergic rhinitis and nasal polyposis were analyzed, providing a research basis on the mechanism of allergic rhinitis and nasal polyps.Materials and methodsTwo samples of inferior turbinate mucosa from cases of nasal polyps and two from cases of allergic rhinitis were collected in this research. The specimens were divided into two parts, one was immersed in10%paraformaldehyde, and with conventional dehydration, paraffin-embedded, then was applied with HE staining for eosinophilic infiltration test. Another was for RNA extraction for further differential expression analysis by RNAseq technology.Result1、ANOVA analysis of variance showed there were significant statistical differences of EOS infiltration among allergic rhinitis, nasal polyps and control group (P<0.01), further S-N-Kq examination showed no statistical difference of EOS infiltrating between allergic rhinitis and nasal polyp group (P=0.670), while EOS infiltration of allergic rhinitis or nasal polyps has a significant statistical difference compared to control group (P<0.01)2、Nasal polyps and allergic rhinitis group shared369differentially expressed genes, with230genes of allergic rhinitis group upregulated at least2times, and139genes downregulated by at least50%compared to nasal polyps group, genes like CXCR4, EGFR, c-fos, and etc were upregulated, involving signaling pathways like MAPK, NOD-like receptors signaling pathways; And other related genes like CIS, ATG1were down-regulated, involving the major signaling pathways like Jak-STAT, mTOR and other pathways. These gens mainly related with the function of cellular growth and proliferation.Conclusions1、Eosinophilic inflammation have played an important role in both allergic rhinitis and nasal polyps pathogenesis.2、Allergic rhinitis and nasal polyps have some relevance in the MAPK, NOD and other signalling transduction pathway.3、Allergic rhinitis is an important factor in the formation of polyps.
