Objective:To observe the effects of erdosteine on activities of superoxide dismutase (SOD), the level of malondialdehyde (MDA) and pathological damage of neurons,the express of BDNF in hippocampus of rats after seizure kindling induced by pentylenetertrazol(PTZ).By this study,inwestigate the effect of erdosteine on the possible mechanism of anti epilepsy.Method:To choose40healthy male SD rats, randomly divided into control,PTZ, ValPr oic acid and erdosteine pretreatment group,10rats in each group.Intraperitoneal injection of PTZ induced epileptic seizure.Erdosteine via stomach for pre-dispo sal treatment before kindling for erdosteine group and gave VPA via stomach f or pre-disposal treatment VPA group.Detected the activities of SOD and the lev el of MDA of right hippocampus; Observed the distribution of BDNF positive cells of left hippocampus by immunohistochemicalstaining; Observed the hippo campal neurons’damages by hematoxylin-eosinstaining (HE).Results:1.The activities of SOD and level of MDA in the hippocampus of rats:The a ctivities of SOD in the hippocampal of epileptic seizure rats,compare tocontrol group, PTZ group were significantly low(p<0.01),and erduosteine group was hig her than PTZ group(p<0.01), no significant difference compared with VPA grou p(p>0.05).level of MDA:In the PTZ group was higher than control group(p<0.01),erdosteine group was lower than PTZ group (p<0.01),and no significant diff erence compared with VPA group(p>0.05).2.The results of HE staining:Neurons in hippocampus of the control group were normal microscopically.The PTZ group which have scattered arrangement of hippocampal neurons,reduction in the number,shrunken of cell body,condensated nuclear appeared a large number of necrotic neurons. There were less neurons in the hippocampus of erdosteine and VPA treatment group. The PTZ group compared control group was significant difference(p<0.01),and compared with erdosteine group was differences(p<0.05).There were no difference between erdosteine group and VPA group(p>0.05).3.BDNF positive cells:The staining intensity in PTZ group increased than control group(p<0.01),but reduced than erdosteine goup(p<0.05). There were no difference between erdosteine group and VPA group(p>0.05).Conclusion:Erdosteine could reduce the oxidative stress level of epileptic rats.lt also inhibit neuronal damage.Erdosteine remove excess free radicals and increased expression of BDNF in hippocampus of PTZ indusced epileptic rats,which may be involved in neuroprotection mechanism.
