ObjectiveIn this study, the AS mouse model for drug intervention, investigate the effect of detoxification method Wenyanghuoxue AS plaque stability from inflammatory immune mechanism. And through clinical studies to explore the therapeutic effect Wenyanghuoxue detoxification method for AMI patients, thus Wenyanghuoxue detoxifying herbs provide evidence based medicine compound of coronary heart disease prevention.MethodsAnimal studies:on100male ApoE-/-mice were given a high fat diet13weeks of modeling, were randomly divided into model group after the model is successful, simvastatin group, low-dose group of Chinese medicine, Chinese medicine dose group and high-dose group.4groups of experimental animals after administration of the drug-related interventions. Drawn13weeks after the intervention, the first mouse serum by ELISA MMP-1, IL-1, IL-8, TNF-α, levels of HIF-1α; then through HE staining, Movat staining finally calculate the plaque vulnerable plaque index; finally assess the impact Wenyanghuoxue detoxifying herbs for AS plaque stability.Clinical Study:40patients with AMI patients met the criteria were randomly divided into experimental Wenyanghuoxue detoxification method Chinese medicine group (+conventional Chinese medicine with western medicine) and the comparison group (simple conventional western medicine). Hs-CRP blood of patients to detect and observe patients before and after administration of TCM symptom score improved in each group, the last joint treatment differences were statistically compare the traditional Chinese medicine with western medicine and western medicine. ResultsExperimental results display:each group of mice after drug intervention, model group and simvastatin group, the serum levels of various inflammatory factors were significantly increased, with significant differences (P<0.05); model group and low dose group There was no significant difference (P>0.05) for each of inflammatory cytokines; model group and the medium dose group of MMP-1, IL-1, no significant difference (P>0.05) TNF-α, HIF-1α levels; model group dose group, increased serum IL-8, with a significant difference (P<0.05); model group and the high-dose group, the serum levels of various inflammatory factors were significantly increased, with significant differences (P0.05); simvastatin group and middle dose group, MMP-1, IL-1, TNF-α, HIF-1α levels were significantly lower, with a significant difference (P<0.05); simvastatin group and the dose IL-8levels were not significantly different (P>0.05); simvastatin group and high-dose groups in traditional Chinese medicine to reduce levels of various inflammatory factors there was no significant difference (P>0.05).Each group of mice after drug intervention, AS plaque vulnerability index overall comparison was significant difference (P<0.05); model group and simvastatin group, the model group was significantly higher vulnerability index, with a significant difference (P<0.05); comparison, no plaque vulnerability index model group and low dose group was significantly different (P>0.05); model group and middle dose group, high dose group, the model group was significantly higher vulnerability index, the difference was statistically significant (P<0.05); simvastatin group, middle dose group and high dose group pairwise comparisons among the three groups, no significant difference (P>0.05).Clinical studies have shown that:before and after treatment, detoxification Wenyanghuoxue Chinese medicine group and the comparison group of patients with hs-CRP were significantly lower symptom scores, with significant difference (P<0.05); Wenyanghuoxue detoxification Chinese medicine and western medicine group compared to the control group, significantly lower symptom scores, with significant difference (P<0.05); Wenyanghuoxue detoxification Chinese medicine group was significantly higher than the total efficiency comparison group, with a significant difference (P<0.05); temperature Yang blood detoxification proprietary Chinese medicine group compared with the control group, serum hs-CRP levels were significantly lower, with a significant difference (P<0.05).ConclusionWenyanghuoxue detoxifying herbs in a stable compound with simvastatin had similar efficacy plaque areas, and are able to drop AS model mice serum levels of inflammatory cytokines have a role in maintaining the stability of vulnerable plaque. Meanwhile, Wenyanghuoxue Chinese medicine detoxification therapy in patients with AMI combined conventional Western medicine, Western medicine treatment is better than medicine to ease symptoms, the total efficiency and reduce blood hs-CRP and other aspects.
