Study On The Effects Of The Model Of The Wenyanghuoxue Detoxification Prescription To Stablize Atherosclerotic Vulnerable Plaque On VEGFs/VEGFRs Singaling Pathway And Clinical Research

ObjectiveThis topic through the mouse models of atherosclerosis drug intervention, to explore this compound of TCM for activating blood detoxification effect on the stability of atherosclerotic vulnerable plaque, and within the plaque angiogenesis mechanism; And through the clinical observation research, explore this huoxue jiedu Chinese medicine compound preparation of traditional Chinese medicine in patients with unstable angina symptoms improvement, thus providing evidence-based medicine for prevention and treatment of coronary heart disease (CHD) of traditional Chinese medicine and other evidence.MethodsAnimal experiment research:of the95male Apoe-/-mice given a high-fat diet13Zhou Zao mould, after the success of the building randomly divided into model group, low-dose group, Chinese medicine dose group, high-dose group and simvastatin group, for4groups of animals were given after drug intervention,13weeks after executed between groups of mice aorta plaque, and rt-pcr method is applied to study the VEGF content in mice aorta change, finally through statistical tests, to determine this compound for the intervention of the vulnerable plaques of TCM for activating blood detoxification.Clinical observation research:meet the conditions for the experiment of30patients with unstable angina were randomly divided into Chinese medicine group (conventional western medicine combined with Chinese traditional medicine) and western medicine group (simple routine western medicine treatment),15cases in each group. Observation group patients of TCM symptom score before and after the treatment, finally through statistical analysis to compare traditional Chinese medicine and western medicine treatment differences.Results1. Plaque vulnerability index, each group of mice after treatment, the overall comparison with significant difference (P<0.05); Multiple comparison between each group, model group and small dose group of plaque vulnerability index was no significant difference (P>0.05); Model group and the middle dose group, high dose group, simvastatin group, model group vulnerability index increased significantly, the difference is statistically significant (P<0.05); Dose group, high-dose group, simvastatin group was no significant difference between the three groups (P>0.05).2. Groups of mice after treatment, model group and low-dose group of Chinese medicine VEGF and VEGFR-2has no obvious difference (P>0.05); Traditional Chinese medicine (TCM) in the dose group, high-dose group, simvastatin group compared with model group, the amount of VEGF and VEGFR-2significantly reduced, statistically significant difference (P<0.05); Traditional Chinese medicine (TCM) in the dose group, the high-dose group and simvastatin group of VEGF and VEGFR-2no difference (P>0.05).3. Clinical observation, the traditional Chinese medicine and western medicine group before and after treatment of TCM symptom score were significantly lower, the difference statistically significant (P<0.05); TCM symptoms curative effect comparison, explored the group total effective rate of TCM for activating blood detoxification than western medicine control group no significant difference (P>0.05).ConclusionA certain concentration of this compound of TCM for activating blood detoxification and simvastatin, can cut atherosclerosis, vascular endothelial growth factor expression in model mice reduce plaque density of new blood vessels, reduce vulnerability index, have a certain role in maintaining the stability of vulnerable plaques. At the same time, this compound with conventional western medicine therapy of TCM for activating blood detoxification patients with unstable angina, can relieve the clinical symptoms.
