Gastric carcinoma is a common digestive system tumor, occurrence ofgastric carcinoma is a multiple steps process that involves many factors andvarious mechanisms, it may be connected with L-selectin and MTDH ofgastric tumor cell.To describe the relationship between L-selectin, MTDH and clinicalpothology characteristics of gastric carcinoma by detecting the expression ofthem in normal gastric mucosa and gastric carcinoma tissue, and to investigatethe role of them in the occurrence and development of gastric cancer, forproviding theoretical basis to evaluate the metastatic potential of malignanttumors and provide theoretical basis for patients prognosis. They may be thenew breach of clinical treatment.Objective:To detect the expression of L-selectin, MTDH protein and mRNA ingastric carcinoma tissue and normal gastric mucosa, analyze the experimentaldata, and then research the relationship between the two and the clinicalpathological characteristics of patients with gastric carcinoma and therelationship of them.Methods:To collect90cases of gastric carcinoma and55cases of normal gastricmucosa (away the tumor more than10centimetres) which were surgicallyresected and confirmed by pathology in many hospitals of Chengde andCangzhou from October2012to March2013. Immunohistochemical SPstaining method was used to detect the expression of L-selectin protein andMTDH protein in normal gastric mucosa and gastric carcinoma tissue; theRT-PCR method was adopted to detect the expression of L-selectin mRNAand MTDH mRNA in the two kinds of tissue. To analyse the experimental data combined clinical pathology with statistics.Results:1The expression of L-selectin protein in gastric carcinoma tissue wassignificantly higher than that in normal gastric mucosa (P<0.05). Theexpression of MTDH protein in gastric carcinoma tissue was significantlyhigher than that in normal gastric mucosa (P<0.05).1.1The positive expression of L-selectin protein in gastric carcinomawhich were poorly differentiated was significantly higher than that in well andmoderately differentiated gastric carcinoma (P<0.05). The positive expressionof L-selectin protein in infiltrated the serosa group was significantly higherthan that in non-infiltrated the serosa group (P<0.05). The expression ofL-selectin protein in lymph node metastasis group was significantly higherthan that of lymph node non-metastasis group (P<0.05). The positiveexpression of L-selectin protein in gastric carcinoma of TNM stage Ⅲ and Ⅳwas significantly higher than that of TNM stage Ⅰand Ⅱ (P<0.05).1.2The positive expression of MTDH protein in gastric carcinoma whichwere poorly differentiated was significantly higher than that in well andmoderately differentiated gastric carcinoma (P<0.05). The positive expression ofMTDH mRNA in gastric carcinoma of TNM stage Ⅲ and Ⅳ was significantlyhigher than that of TNM stageⅠand Ⅱ (P<0.05).There were no relationships between L-selectin mRNA, MTDH mRNAand every other characteristics of gastric carcinoma such as age, gender, typeof pathology (P>0.05, P>0.05, P>0.05).3The relativity of their expression:3.1The correlation coefficient of L-selectin protein and MTDH protein is:r=0.341(P<0.05).3.2The correlation coefficient of L-selectin mRNA and MTDH mRNA is:r=0.369(P<0.05).Conclusions:1Immunohistochemical positive signals of L-selectin protein and MTDHprotein were significant differences between normal gastric mucosa andgastric carcinoma, also were concerned with clinical pathology characteristics(differentiation degree, invasive depth, metastasis of lymph nodes, TNM staging of gastric carcinoma).2L-selectin mRNA and MTDH mRNA were significant differencesbetween normal gastric mucosa and gastric carcinoma, also were concernedwith clinical pathology characteristics(differentiation degree, invasive depth,metastasis of lymph nodes, TNM staging of gastric carcinoma).3The correlation of L-selectin and MTDH was positive; they may playpositive cooperativity in the occurrence and development of gastriccarcinoma.
