Objective:To explore the expression of IGF-II、IGF-IR and IGFBP-6in theoccurrence and development of human colorectal carcinoma by detecting theexpression of these factors in normal mucosa, colorectal adenoma andcolorectal carcinoma,and to analyze the interaction of IGF-II、IGF-IR andIGFBP-6and the relationship of these factors with the clinical pathologicalcharacteristic of colorectal cancer patients such as age,gender,tumorlocation,depth of invasion, lymph node metastasis,and Duke’s stage so as toevaluate the relationship among these factors.Method:50cases of colorectal carcinoma (CC),20cases of colorectal adenoma(Ad) and50cases of colorectal normal mucosa(NM) were collected from Jan,2012to Dec,2012, from Surgical Oncology of Chengde Medical Collegeaffiliated hospital.All specimens were surgically removed and certified bypathology. Immunohistochemical and RT-PCR methods was adopted toexamine the expression of IGF-II、 IGF-IR and IGFBP-6in colorectalcarcinomas(CC),adenoma(Ad) and normal tissues(NM).Results:1The expression of IGF-II mRNA,、IGF-IR mRNA and IGFBP-6mRNA,IGF-II、IGF-IR and IGFBP-6in normal mucosa, colorectal adenoma andcolorectal carcinoma.1.1The expression of IGF-IImRNA、IGF-IR mRNA and IGFBP-6mRNA inNM, Ad, CCIGF-IIand IGF-IR mRNA in CC and Ad was significantly higher thanthat in NM (P<0.05、P<0.05、P<0.05、P<0.05),and in CC was significantly higher than that in Ad (P<0.05、P<0.05). IGFBP-6mRNA in Ad and CC wassignificantly lower than that in NM (P<0.05、P<0.05), and in CC wassignificantly lower than that in Ad (P<0.05).1.2The expression of IGF-II, IGF-IR and IGFBP-6in NM, Ad, CCThe expression of IGF-II in CC and Ad was significantly higher thanthose of in NM (P<0.05、P<0.05). The expression of IGF-IR in CC and Adwas significantly higher than those of in NM (P<0.05、P<0.05), and in CCwas significantly higher than that in Ad (P<0.05).The expression of IGFBP-6in CC and Ad was significantly lower than those of in NM (P<0.05、P<0.05),and in CC was significantly lower than that in Ad (P<0.05).2The relationship between IGF-II, IGF-IR and IGFBP-6and clinicalpathological characteristic of colorectal carcinoma.The expression of IGF-II and IGF-IR were positively correlated withdepth of invasion、lymph node metastasis and Duke’s stage. The expression ofIGF-IIand IGF-IR in CC of infiltration more than musucularis wassignificantly higher than those not (P<0.05, P<0.05);lymph node metastasisgroup was significantly higher than those without lymph nodemetastasis group(P<0.05, P<0.05);Duke’s C+D stage was significantly higher than Duke’s A+B stage(P<0.05, P<0.05).The expression of IGFBP-6in CC of infiltrationmore than musucularis was significantly lower than those not (P<0.05);lymphnode metastasis group was significantly lower than those without lymphnodemetastasis group (P<0.05);Duke’s C+D stage was significantly lowerthan Duke’s A+B stage (P<0.05).The expression of IGF-II、IGF-IR andIGFBP-6were not found correlations with every other characteristics of CCsuch as age、gender、differentiation and tumor location(P all greater than0.05).3The relationship among IGF-II、IGF-IR and IGFBP-6The positive correlation could be seen between IGF-II and IGF-IR (r=0.59, P<0.05);The negative correlation could be seen between IGF-II andIGFBP-6(r=-0.61, P<0.05);The negative correlation could be seen betweenIGF-I R and IGFBP-6(r=-0.75, P<0.05). Conclusions:1The expression of IGF-II、IGF-IR and IGFBP-6may play an importantrole in the occurrence and development of CC. IGF-II、IGF-IR may promotethe development of CC while IGFBP-6Inhibition it.2IGF-II、IGF-IR and IGFBP-6was significantly correlated with depth ofinvasion,lymph node metastasis and Duke’s stage in CC. The result suggestedthat these factors can be used as a predictive factor for CC.3There were significantly correlation among IGF-II、 IGF-IR andIGFBP-6in CC. High expression of IGF-II in colorectal carcinoma, IGF-IRhigh expression, the combination of synergistic effect on occurrence anddevelopment of CC,both play the role of promote the development of CC.When IGFBP-6high expression in colorectal carcinoma, IGF-II and IGF-IRlower expression, able to predict the high expression of IGFBP-6can inhibitthe action of IGF-II and IGF-IR.It can be predicted by some way to inhibit orblock the expression of IGF-II and IGF-IR, or increase the expression ofIGFBP-6may reduce the development and transfer of CC.
