Objective: To investigate the expressions of hypoxia inducible factor 2 alpha (HIF-2α),CC chemokine receptor 7(CCR7)and vascular endothelial growth factor C(VEGF-C) as well as the relationships among them in human colon carcinomaMethods: HIF-2α, CCR7 and VEGF-C expression of 60 cases resected specimens of colon cancer and 20 cases specimens of normal colon tissues were detected using SABC immunohistochemical methods. Correlations among them and their relationships with clinicopathologic characteristics of human colon cancer were analyzed byχ~2 test or Spearman analysis or Wilcoxon rank sum test.Results:①The percentage of HIF-2αpositive expression is 71.67% in colon cancer(43/60), but none positive expression was found in normal colon tissues(0/20). and significant differences were observed between them(p﹤0.01); The percentage of CCR7 positive expression is 81.67% in colon cancer(49/60), but the percentage was 5.00% in normal colon tissues(1/20). and significant differences were observed between them(p﹤0.01); The percentage of VEGF-C positive expression is 75.00% in colon cancer(45/60), but the percentage was 5.00% in normal colon tissues(1/20). and significant differences were observed between them(p﹤0.01);②The positive expressions of HIF-2αand CCR7 in colon cancer were closely associated with lymph node metastasis, invasive depth, and Dukes stages(p﹤0.05), the grades of HIF-2αand CCR7 positive expression had no significant differences in groups with lymph node metastasis, invasive depth, and Dukes stages(p﹥0.05), but it had no association with gender, ages, tumor diameter, and tumor differentiation grade(p﹥0.05); The positive expression of VEGF-C in colon cancer was closely associated with tumor differentiation grade, lymph node metastasis, invasive depth, and Dukes stages(p﹤0.05), the grades of VEGF-C positive expression had no significant differences in groups with tumor differentiation grade, lymph node metastasis, invasive depth, and Dukes stages(p﹥0.05), but it had no association with gender, ages and tumor diameter(p﹥0.05).③Significant correlations were observed between HIF-2αand CCR7 in colon cancer(rho=0.786,p﹤0.01); Significant correlations were observed between HIF-2αand VEGF-C in colon cancer(rho=0.695,p﹤0.01); Significant correlations were observed between CCR7 and VEGF-C in colon cancer(rho=0.858,p﹤0.01).Conclusions: 1. HIF-2α, CCR7 and VEGF-C were all highly expressed in colon cancer tissue; 2. Positive expression of HIF-2α, CCR7 and VEGF-C in colon cancer was correlated with invasive depth, lymph node metastasis and Dukes stages, furthermore Positive expression of VEGF-C has negative correlation with tumor differentiation grade; 3. Positive correlations were observed between HIF-2αand VEGF-C, CCR7 and VEGF-C, HIF-2αand CCR7 in colon cancer; 4. The detection of HIF-2α, CCR7 and VEGF-C in colon cancer is a indicator to assess its prognosis.
