Objective: Observe the effects of traditional Chinese medicine preparation Tongmai pill on thenumber and function of peripheral blood progenitor cells in rat’s with arteriosclerosis obliterans.Methods:1To establish arterial occlusion modelExperiments using SD rats for the study, the weight of165.6g±19.4gSD60malerats were randomly divided into30groups, making the module30.All modules will bemade ligation and transection double rat femoral artery branch, and with toothedforceps holding approximately30s, monthly intramuscular vitamin D3300thoundsu/kg once.After8weeks, two groups of rats were randomly selected, testin gserum TC,TG, LDL-C, HDL-C content and femoral artery biopsy.2Effects of Tongmai Pill on numbers and function of EPCsThe successful modeling24ASO rats were randomly divided into four groups,the control group and the low, medium and high dose group (1.5g/kg,5/kg,9g/kg),were treated with different concentrations tongmai pill concentrate gavage the controlgroup with the same volume of saline, blood was centrifuged to obtain after7daystongmai pills containing serum.SD rat aorta pumping blood by density gradient centrifugation charged withmononuclear cells, adding VEGF and bFGF induced differentiation, culture7d aftercollecting adherent cells. FITC-UEA-1and Dil-ac-LDL double positive cells wereidentified as EPCs, with CCK-8, Tanswell chamber were detected EPCs proliferation,adhesion, migration and with the statistical analysis. Results:1.Two groups of rats fed different diets for8weeks no significant difference in bodyweight, rat femoral artery lumen model group after modeling stenosis, thrombosis, intimal structuralabnormalities, medial smooth muscle proliferation and subcutaneous inward migration, internalelastic board damage, loss of endothelial cells, connective tissue disorder, visible foam cells in theplaque.2.Tongmai pill containing serum ASO can increase the number of male rats EPCs affect serumcontaining medium dose group (5g/kg) tongmai pill after the intervention of EPCs number of themost significant (P<0.05), significantly tongmai pill EPCs improved proliferation, adhesion andmigration capabilities.Conclusion:1.Arteriosclerosis obliterans model can be successfully replicated;2.Tongmai pillscan increase the number of ASO rat’s EPCs and improve its functionality.
