Effects Of Blood Glucose Fluctuation In Diabetic Rats Of Kidney Disease And Oxidative Stress

Objective:To explore the effects of blood glucose fluctuation of diabeticrats of kidney disease and oxidative stress.Methods: SD rats were divided into4groups:control group (group A)、normal blood glucose fluctuation group(group B)、diabetic stable high blood glucose group(group C) and fluctuanthigh blood glucose group (group D). Diabetes rats were induced byintraperitoneal injection of STZ，the fluctuant high blood glucose animal modelwas induced by subcutaneous injection insulin and intragastric administrationglucose at different time points every day.9weeks later to detect the bloodglucose, creatinine, urea nitrogen, glycosylated hemoglobin in blood samplingand to caculate SDBG、LAGE,、MBG and to detect the microalbuminuria,Theactivities of antioxidant enzymes in serum, including the total superoxidedismutase (TSOD) glutathione peroxidase (GSH-Px) and catalase (CAT),malondialdehyde (MDA) were measured by chromatometry and to observe thechange of kidney microstructure by optical microscope.Results: there was noSignificant differences between A group and B group.the C、D groupcompared with the A、B group: Significant weight reduce,and Significantincrease Of fasting blood-glucose（FBG）、SDBG、LAGE、MBG、Cr、SUN、 HbA1c and MAG,the activity of TSOD、GSH-Px、CAT were decreasedsignificantly,but the increase of MDA,Inspection of the renal tissue by opticalmicroscope demonstrated that the increase volume and the expansion or ruptureof glomerulus, reduce of glomerulus cells and the infiltration of Inflammatorycells, renal tubular structure disorder compared with C group,there’s moresignificant changes in D group.Conclusion: blood glucose fluctuation canaggravate diabetic rats of kidney disease and oxidative stress and play animportant role in the pathogenesis of diabetic nephropathy.
