Radix Rehmanniae Praeparata, flavour kenndey and sex lukewarm, distribute to liver andkidney channel, efficacy nourishing blood and raise energy. And can be used to treat multiplesymptoms, such as hemopenia, lack of essence, liver and kidney yin deficiency. Widely applied inthe clinical medicine. Almost1/10contents of the traditional Chinese medicine prescriptionpreparation recorded by the2010edition of Chinese Pharmacopoeia involved Radix RehmanniaePraeparata.Radix Rehmanniae Praeparata is made by steamed or wine stew fresh or dried tuberous rootof scrophulariaceae plant Rmannia. Annotation of Materia Medica and Lie Gongs Treatise onPrepatation and Boiling of Materia Medica the earlieat dovumented the stesmed and wine stemedmethod of Radix Rehmanniae Praeparata. Only after a long process we can obtain the sweet andblacd Radix Rehmanniae Praeparata. During this process, the sex of Rehmannia change fromlukewarm to cold. Although distribute to liver and kidney channel, but the efficacy change fromeclear away heat catambola and cooling blood to stop bleeding to mourishing blood and raiseenergy, and then becaome the preferren medicina materials to raise blood deficiency.Radix Rehmanniae Praeparata contains a variety of chemical composition such as glycosides,scccharide, amino acids, metal ion, and so on. Now, many academics believe that the iridoidglycosides is the principal effective comopents of Rehmanniae. New study has found that iridoidglysocides Rehmannioside A Rehmannioside D relatively stable compared with the othercomposition in Rehmanniae, which can be used as the major components to control the quality ofRehmannia. We start from extract the main iridoid glysocides effective components of Rehmanniae:Separation, refining and purification the fresh Rehmannia which extract by methanol, using themethods, such as macroporous adsorption resin,silica gel column chromatography, glucan gel,efficient preparation of liquid phase. Final got four kinds of iridoid glysocides standard materialfrom Rehmannia: Onuride, Rehmannioside A and Rehmannioside D. and their purity were over98%. which measured by the high performance liquid chromatography (HPLC).We research the stsbility of the three standard material of Rehmanniae: Catalpol;Rehmannioside A and Rehmannioside D in different envuronments on the foundation of gotstandard materials of Rehmanniae. Using the high performance liquid chromatography (HPLC)determined the HPLC figure of Catalpol; Rehmannioside A and Rehmannioside D in differentenvuronments, by comparing the change of the chromatographic pesks, explore its stability.Results, Ultraviolet light had the worst influence on the stabilith of catalpol, Rehmannioside A andRehmannioside D. Followed by acid; alkali; oxidant. Moreor, reducing agent; high temperature;Na+, Cu2+, Fe3+, Ag+, Ca2+metal ions also have certain influence to Catalpol. So,we should avoidthe environment which cold cause the sctive ingredients of radix rehmanniae degradation, to makeit play a better effect.We establish the quality evaluation standards of Radix Rehmanniae Praeparata. Make thequalitative identification of Rehmannioside A, Rehmannioside D and Acteoside in differentsamples by the thin layer chromatography (TLC); make the content determination ofRehmannioside A, Rehmannioside D and Acteoside in different samples by the reverse phase highperformance liquid chromatograph (RP-HPLC). Results: TLC have the better specificity anddistinctive feature; The Rehmannioside A showed a good linearity with the rang of0.296~1.776 μg, Y=1.99916*10-6x-0.0267363(r=0.9997); Rehmannioside D showed a good linearity with therang of0.684~3.42μg, Y=2.65844*10-6x-0.0456232(r=0.9996); and Acteoside resectivelyshowed a good linearity with the rang of0.0816~0.408μg, Y=5.28041*10-6x-0.0229814(r=0.9998), which assaying by HPLC. The established method is rapid, convenient and accurate,which can be used to control the quality of Radix Rehmanniae Praeparata.Extract the main effective components, research the stsbility of Rehmanniae and establish thequality standard of Radix Rehmanniae Praeparata which can provide the basis for the reasonableand effective use of Rehmanniae, more further lay the foundation to development and studiesRehmannia.
