| Objective The Kruppel-Like Factors5(KLF5) gene, sites at chromosome13q21.3, Including18.72kb region, NCBI No.688, starts at73633142and ends at73651676bp, KLF5has a close relationship with the occurrence of breast cancer development.This study by detecting the expression of KLF5in breast cancer tissue〠adjacent tissue and breast adenosis tissue, and analysis the correlation between expression level of KLF5in breast cancer and clinical characteristics. Preliminary clarify KLF5’s role in the development of breast cancer, and further explore its relationship with triple negative breast cancer.Methods The expression of KLF5in100cases of breast cancer tissuesã€100cases of adjacent tissues and20cases of breast adenosis tissues was determined by immunohistochemistry.(All the cases were female. Pathological type:invasive ductal carcinoma57cases, invasive lobular carcinoma13cases and Carcinoma in situ11cases. Lymph node metastasis:Axillary lymph node metastasis in56cases, and44cases were found no axillary lymph node metastasis. Pathological grade:The grade â… +â…¡ contained53cases. The grade â…¢+â…£ contained47cases. The group which below50-years-old was47cases and above50-years-old was53cases. The group with tumor thrombosis was12cases without was88cases. The tumor size larger than5cm was10cases but smaller than5cm was90cases. ER:Positive,65cases. Negative,35cases. PR:Positive,68cases. Negative,32cases. C-erbB-2:Positive,23cases. Negative,77cases. We analyzed the relationship between the expression level of KLF5and pathological characteristics such as pathological typeã€lymph node metastasisã€histological grade〠ageã€tumor sizeã€vascular or withoutã€estrogen receptor (ER) and progesterone receptor (PR)ã€C-erb-2ã€P170and so on.Results The expression of KLF5is mainly localized in the cytoplasm. KLF5proteins can see the positive expression in breast cancer tissue and in para-carcinoma tissues and breast adenosis tissues,however, KLF5protein expression in breast cancer cells was significantly higher than the in para-carcinoma tissues and breast adenosis tissues (x2=19.003, P=0.000). The number of high level expression in breast cancer tissue was74cases (74/100,74.0%), and the number of low level expression or absent was26cases (26/100,26.0%); The number of high level expression in para-carcinoma tissue was48cases (48/100,48.0%), and the number of low level expression or absent was52cases (52/100,52.0%); The number of high level expression in breast adenosis tissues was7cases (7/20,35.0%), and the number of low level expression or absent was13cases (13/20,65.0%)。100cases of breast cancer, KLF5positive expression rate was74.0%(74/100,74.0%), significantly higher than the para-carcinoma tissue48.0%(48/100,48.0%)(P<0.05) and adenosis tissues35.0%(7/20,35.0%)(P<0.05), while the comparison between para-carcinoma tissue and adenosis tissues was no statistically significant (P>0.05). The number of high expression of KLF5was38(38/57,66.7%) and the low or absent was19(19/57, 33.3%) in group of invasive ductal carcinoma, the numbers were7(7/13,53.8%) and7(6/13,46.2%) in invasive lobular carcinoma and the numbers were10(10/11,90.9%) and1(1/11,9.1%) in carcinoma in situ (x2=4.419, P=0.110). The number of high expression of KLF5was42(42/56,75.0%) and the low level expression or absent was14cases (14/56,25.0%) in group with axillary lymph nodes metastasis, however, in group without axillary lymph nodes metastasis the numbers were32(32/44,72.7%) and12(12/44,27.3%)(x2=0.066, P=0.797). The number of high expression of KLF5was43(43/53,81.1%) and the low level expression or absent was10cases (10/53,18.9%) in group with pathological grade â… +â…¡, however, in group grade â…¢+â…£ the numbers were31(31/47,66.0%) and16(16/47,34.0%)(x2=2.981, P=0.084). The number of high expression of KLF5was31(31/47,66.0%) and the low level expression or absent was16cases (16/47,34.0%) in group with age below50-years-old, however, in group with age above50-years-old the numbers were33(33/53,62.3%) and20(20/53,37.7%)(x2=0.147, P=0.701). The number of high expression of KLF5was10(10/12,83.3%) and the low level expression or absent was2cases(2/12,16.7%)in group with tumor thrombosis, however, in group with no tumor thrombosis the numbers were64(64/88,72.7%) and24(24/88,27.3%)(x2=0.617, P=0.432). The number of high expression of KLF5was64(64/90,71.1%) and the low level expression or absent was26cases (26/90,28.9%)in group with tumor size below5cm, however, in group with tumor size above5cm the numbers were10(10/10,100.0%) and0(0/10,0.0%)(x2=3.904, P=0.048). The number of high expression of KLF5was50(50/65,76.9%) and the low level expression or absent was15cases (15/65,23.1%) in group of ER(+), however, in group of ER(-) the numbers were24(24/35,68.6%) and11(11/35,31.4%)(x2=0.825, P=0.364). The number of high expression of KLF5was52(52/68,76.5%) and the low level expression or absent was16cases (16/68,23.5%) in group of PR(+), however, in group of PR(-) the numbers were22(22/32,68.7%) and10(10/32,31.3%)(x2=0.674, P=0.412). The number of high expression of KLF5was15(15/23,65.2%) and the low level expression or absent was8cases (8/23,34.8%) in group of C-erbB-2(+), however, in group of C-erbB-2(-) the numbers were59(59/77,72.6%) and18(18/77,27.4%)(x2=1.198, P=0.274). The number of high expression of KLF5was45(45/60,75.0%) and the low level expression or absent was15cases (15/60,25.0%) in group of P170(+), however, in group of P170(-) the numbers were29(29/40,72.5%) and11(11/40,27.5%)(x2=0.078, P=0.780). The number of high expression of KLF5was11(11/14,78.6%) and the low level expression or absent was3cases (3/14,21.4%) in group of triple negative breast cancer, however, in group of no triple negative breast cancer the numbers were63(63/86,73.3%) and23(23/86,26.7%)(x2=0.177, P=0.674). The data shows that the level of KLF5expression has relationship with breast cancer tissueã€adjacent tissueã€breast adenosis tissue and tumor size, but has no relationship with pathological typeã€lymph node metastasis〠histological gradeã€ageã€tumor sizeã€vascular or withoutã€estrogen receptor (ER) and progesterone receptor (PR)ã€C-erb-2ã€P170and so on.Conclusion The expression of Zinc finger protein transcription factor5gene is mainly localized in the cytoplasm. The different degrees of expression of KLF5in breast cancer tissueã€para-carcinoma tissues and adenosis tissues, but KLF5protein expression in breast cancer cells was significantly higher than the in para-carcinoma tissues and breast adenosis tissues. Upregulation of its expression may be intensity associated with the occurrence of breast cancer and development. The expression level of KLF5related to tumor size, equal to or greater than the5cm tumor tissue the expression of KLF5significantly higher than the less than5cm organization. It has no relationship with pathological typeã€lymph node metastasis〠histological grade〠ageã€tumor sizeã€vascular or withoutã€estrogen receptor (ER) and progesterone receptor (PR)ã€C-erb-2ã€P170and so on. Detecting KLF5expression in breast cancer may become important marker to judge staging of breast cancer. |
