Abstract The fermentation substrates (such as corncobs, bagasse, wheat bran, rice bran and so on) mixed with mycelium, substrates and secondary metabolites are called the culture of hericium erinaceus. The culture of hericium erinaceus is produced by bidirectional solid fermentation technology. The culture of hericium erinaceus has a curative effect on the gastrointestinal disease. In order to clearly understanding the material basis on gastrointestinal disease and relevant pharmacological activities of the culture of hericium erinaceus, we test the pharmacological activity of the culture and the chemical constituents for the first time.We firstly validated the anti-helicobacter pylori and antitumor activity, which determined by the effective part of the culture of Hericium erinaceu. By50%ethanol reflux extraction, flash type extraction at room temperature, and some different process, we got seven different parts. We found that part-2has the best resistant activity of helicobacter pylori and inhibition activity of the SW620colon cancer cell growth by in vitro anti-helicobacter pylori experiment and antitumor MTT method.Eighteen compounds were seperated from the effective part-2of the culture of hericium erinaceus, including two flavonolignans, six flavonoids, four steroidals, five anthraquinones and one alkaloid; (12R,13R)-Flavonolignan A is new compound, All compounds were firstly isolated from the culture of Hericium erinaceus.We firstly screened the active ingredients of anti-helicobacter pylori and antitumor activity. The experimental results showed that all the five anthraquinone compounds have anti-helicobacter pylori activity, and emodin and aloeemodin have the strongest anti-helicobacter pylori activity in vitro. We also screened schaftoside, isoliquiritigenin, tricin, salcolin and (12R,13R)-Flavonolignan A with the antitumor activity by MTT method, The experimental results showed that the five components have a certain inhibitory effect on HepG2liver cancer cells and Caco-2colon cancer cells. Schaftoside had the best inhibitory effect on HepG2liver cancer cells; tricin and (12R,13R)-Flavonolignan A showed the best inhibitory effect on Caco-2bowel cancer cells.Our reseach provided the theoretical basis for pharmacological mechanism reseach of the Hericium erinaceus culture, and it also can provide some experience reference for bi-directional solid fermentation of medicinal fungi.
