Hericium erinaceus is a famous edible and medicinal fungi. The mushroom has a sweet taste and mild pharmacological property. It has been widely proved that it is effective in curing many diseases. It possesses the function of helping the digestion and beneficial to internal organs. It shows a good effectiveness on various diseases of digestive tract, such as the duodenum ulcer and gastric ulcer. Every 100 grams of dried fungus contain 26.3 g protein, 4.2 g fat. Hericium erinaceus polysaccharide is the major active component in the fungi. In this research, many works have been done about hericium erinaceus polysaccharide (HEP) as the following. We try to isolate the exact substance that cure gastric ulcer from the liquid-culture. It should be important to make further study on the medicinal active compounds from hericium erinaceus. For the reason, we compare the curative effect of HEP to the Weilexin and the culture of hericium erinaceus removed HEP. In addition, we have done a lot of detail research on the HEP-2 that is separated from HEP by use of Sephadex-G150.To begin with, the liquid-culture was deluted by distilled water, then the ethanol was added to the culture solution till the deepness of ethanol was about 80%. The polysaccharide was deposited from the culture of mycelia of hericium erinaceus. The deposition was dried and named section one whose composition mainly was HEP. The left culture solution was named section two containing many low molecular weight organic compounds such as simple sugar, fatty acid, sterol etc. Sixty Wistar mice were divided into six groups and used for the model of experiment gastric ulcer. Selecting 10 normal Wistar rat as control group, and 50 above mentioned rat which were divided into five groups such as: gastric ulcer positive group ,weilexin group (6.45g/kg), section one group(0.0542g/kg) , section one group(0.0271g/kg) , section two4group(1.88g/kg) , section two group(0.94g/kg) .Each group has ten. The result was gained from the assay and method of gastric ulcer area and it showed that the section one was more effective on the gastric ulcer.Secondly, the study was focused on HEP. The section one (HEP) was composed of polysaccharide, protein and mineral. The content of polysaccharide was determined by use of phenol- sulfuric acid method. That of the protein was determined by use of Lowry method. The results showed HEP contained 31.7% polysaccharide and 10.6% protein.The protein of HEP was got rid of by Sevag method. Then, HEP was separated from the Sephadex-Gl50.The HEP-1 and HEP-2 were gained from the eluted solution. HEP-2 was dried and determined 62.5% polysaccharide, 7.5% protein and no uronic acids. The composition and structure of HEP were concluded from the following data:The purity and molecular weight of HEP-2 were determined by HPGPC and Sephadex G150.The result of GC showed that HEP-2 was composed with Ara, Man, Glu and the proportion was Ara:Man:Glu = 1: 6.28: 22.96.HEP was hydrolyzed and partial methylated and partial acetated. The partial methylated derivation was analyzed by GC-MS. The result showed that HEP-2 contained eleven methylated alditol: 2,3,4,6-Me4-Glc, 3,5-Me2-Ara, 2,3,6-Me3-Man, 2,3,4-Me3-Glc, 2,3-Me2-Glc, 2,4-Me2-Glc, 2-Me-Glc, 3-Me-Man, 3-Me-Glc, 6-Me-Man, 6-Me-Glc. The molar ratio was 135.: 1: 9.22: 6,62: 3.84: 0.60: 0.67: 0.12: 0.12: 0.17: 0.36. The main chain of HEP-2 was -l)Man(4- and -l)Glc(6-. The chain's end was mainly -l)Glc. The kinds of fatty acids in HEP-2 were methyled and detected by GC-MS. The contents of fatty acids were determined by GC. The results showed that HEP was consisted of Tetradecanoate, 9-Hexadecenoic acid, Hexadecanoic acid, 8,11-Octadecadienoic acid, 8-Octadecenoic acid, Octadecanoic acid, but thequantity was trace. The result of EA showed that HEP was consisted of C 27.47%, H 4.08%, N 1.24%, O 47.83%, S 0.48% and that of ICP-AES shows that there are many mineral elements in HEP.
