Objective:To investigate the the expression of the novel estrogen receptor GPR30in mRNA and protein level of placenta and the relationship between the expression of GPR30with TBA, estradiol(E2) level in serum and the prognosis of the fetus in patients of intrahepatic cholestasis of pregnancy (ICP), speculating the possible role of GPR30in the placenta of ICP patients and the influence to the fetus.Methods:Peripheral serum and placenta tissues were obtained from30patients with ICP(15with mild ICP,15with severe ICP,depending on the levels of TBA in the Peripheral serum) and15normal pregnant women (NP group).The concentrations of E2in the peripheral serum was tested by ELISA, and real-time quantitative polymerase chain reaction and Western blot were used to eavluate the expression of GPR30mRNA and protein in placenta tissues.The appearance of the factors led to the poor prognosis of fetus is collected.Results:1. The serum TBA concentration of the severe ICP group was (48.51±8.39) μmol/L, while the mild ICP group was (22.04±7.11) μmol/L,and the normal pregnant group was (3.42±1.31) μmol/L; pairwise comparisons were statistically significant (P<0.01).2.The serum E2concentration of the severe ICP group was (33.58±1.70) ng/ml,while the mild ICP group was (28.66±1.66) ng/ml,and the normal pregnant group was (23.83±1.16) ng/ml; pairwise comparisons were statistically significant (P<0.01).3.The relative expression of GPR30mRNA in placenta tissues of the severe ICP group was (2.78±0.53),while the mild ICP group was (1.99±0.27),and the normal pregnant group was (1.27±0.24); pairwise comparisons were statistically significant (P<0.01).4.The expression of GPR30protein in placenta tissues of the severe ICP group was (0.54±0.09),while the mild ICP group was (0.40±0.55),and the normal pregnant group was (0.32±0.04); pairwise comparisons were statistically significant (P<0.01).5.There is a positive correlation between the relative expression of GPR30mRNA with E2and TBA,the correlation coefficient "r"=0.812and0.819;6.There is also a positive correlation between the expression of GPR30protein with E2and TBA, r=0.741and0.796.7. There is a positive correlation between the relative expression of GPR30mRNA with the expression of GPR30protein,r=0.829.8. The patients with poor prognosis of fetus had a higher relative expression of GPR30mRNA in placenta tissues (2.41±0.531) than the others (1.69±0.71); they also had a higher expression of GPR30protein in placenta tissues (0.48±0.10) than the others (0.35±0.12)(P<0.01)。Conclusions:The E2concentration in serum is elevated in ICP patients and is related to the severity of the disease.GPR30is expressed in human placenta, The relative expression of GPR30mRNA and the expression of GPR30protein are elevated in ICP patients compare with the normal pregnancy, and they are positively correlated with the TBA level, providing a clue that GPR30is associated with the severity of ICP.The relative expression of GPR30mRNA and the expression of GPR30protein both have a positive correlation with E2concentration, the interaction between GPR30and E2may contribute to the placenta’s sensibility to E2. The high expression of GPR30protein is associated with the poor prognosis of the fetus,this is presumedly caused by E2,which is the ligand of GPR30and make the transportion of TBA between maternal and fetal blood out of balance.
