Abstract:Houttuynia cordata Thunb.(HCT), a perennial plant of Houttuynia of Saururaceae, is a well-known traditional Chinese medicine widely used in China. It possesses functions of clearing heat, eliminating toxins, reducing swelling, discharging pus, relieving cough and resolving phlegm. In the present study, the quality control of Houttuynia cordata and metabolism of volatile active ingredient methyl nonyl ketone in rat were systematically investigated by gas chromatography-mass spectrometry (GC-MS). The results will provide a scientific basis for quality control and clinical application of Houttuynia cordata.Firstly, this study analyzed the volatile components of Houttuynia cordata under different light intensity by gas chromatograph-mass spectrometer (GC-MS) combined with chemometric methods. There were some similarities and differences in volatile oil fingerprint among3groups under different light intensities. Totally,33compounds were identified in the volatile oil from3groups, of which26components were common to all materials. The content of decanoyl acetaldehyde and2-undecanone in group with full light were significantly lower than two groups with shading. The results suggested that moderate shading was beneficial to the improvement of content of effective components of Houttuynia cordata Thunb volatile. The types and content of volatile components from Houttuynia cordata Thunb was closely related to the light conditions.Secondly, a rapid, stable, sensitive and accurate method for determination of methyl nonyl ketone in rat plasma and tissues at different times after oral administration was developed based on GS-MS. The concentration-time curves showed that methyl nonyl ketone was absorbed rapidly and distributed widely in rat plasma and various tissues, followed by quick elimination, and had no long-term accumulation in vivo.Lastly, rat plasma endogenous metabolites in blank and medicine groups of different dosage were analyzed after deproteinization and derivatization which includes oximation and silanization. Totally,43metabolites were identified in both blank and medicine groups. Among them,3metabolites showed significant difference in content of blank and medicine groups. The results suggested that methyl nonyl ketone has an influence on the metabolism of rats.
