Houttuynia cordata Thunb. is a Chinese materia medica that is generally used in the Chinese medicine therapy. It possesses the actions of clearing heal eliminating toxins, reducing swelling, discharging pus and relieving stagnation. To make the most of it, Houttuynia cordata Thunb. was studied comprehensively in our research. On the one hand, Flavonoids which are the main active ingredients of Houttuynia cordata Thunb. were purified to provide raw material for the development of new drugs. On the other hand, HPLC-DAD fingerprint and GC fingerprint were established to reflect the chemical composition completely.The first fraction of this thesis was to separate and purify flavonoids (chapter2). The organic solvent extraction, the silica gel column chromatography, high performance liquid chromatography, along with semi-preparative liquid chromatography were used to get monomers of flavonoids. Finally,6monomers of flavonoids were purified. Compared with the standard substance and information obtained from LC-MS,4monomers were identified to be quercitrin, quercetin, hyperin and isoquercitrin. The purity of4kinds of substances counted was98.31%,96.11%,99.25%and89.7%by chromatographic peak area normalization. The second fraction of this thesis focused on non-volatile components analysis and volatile components analysis with high performance liquid chromatography-diode array detector (HPLC-DAD) fingerprint and GC (gas chromatography) fingerprint (chapter3and chapter4). Houttuynia cordata Thunb. are grown widely and orgin will impact TCMs’ active ingredients greatly. So, the non-volatile components of Houttuynia cordata Thunb. from12different localities with HPLC-DAD method were analyzed (chapter3). The content included optimization of extraction, selection of suitable chromatographic separation conditions, validation of methodology, similarity analysis and the main active ingredients content analysis. The results showed that the fingerprints of non-volatile constituents were very similar. The similarity was greater than0.92. But the quantitative research of three main active compounds showed that the content of different samples was very different. Meanwhile, non-volatile components in Houttuynia cordata Thunb. were researched during storage. The storage study showd that non-volatile components in Houttuynia cordata Thunb. were stable in12monthes.In chapter4, volatile components of Houttuynia cordata Thunb. from7different localities were determined with GC method. Correlative similarity comparative analysis (SCA) combined with quantitative analysis was used to study Houttuynia cordata Thunb.. The result demonstrated that the fingerprints were similar considerably, but the content of main active ingredients in different samples was very different. The result was the same as non-volatile components analysis. Therefore, as the medicinal use of Houttuynia cordata Thunb., the origin of the herbs should be taken into consideration seriously. The combination of HPLC-DAD fingerprint and GC fingerprint would control the quality of Houttuynia cordata Thunb. comprehensively.
