Objective: To study the role of miR-30a in bone morphogenetic protein9(BMP9) induced osteogenic differentiation of mesenchymal stem cell lineC3H10T1/2.Method: C3H10T1/2cells were treated with Adenovirus GFP andBMP9, the early osteogenic marker Alkaline phosphatase(ALP) activitywas detected by ALP quantitative assay and staining assay, the laterosteogenic marker calcium precipitation was detected by Alizarin Red Sstaining,OPN was another osteogenic marker, which was detected by PCRand Western blot. The expression of miR-30a in C3H10T1/2cellsquantified by Real-time PCR. Effects of miR-30a on BMP9-inducedosteogenic differentiation of C3H10T1/2cells was detected by ALP, OPNand calcium precipitation. Cell proliferation analysis was determined byMTT and the flow cytometry. Runx2was detected in order to study itsmechanism.Results: BMP9was not only increased the activity of ALP on days7, the calcium precipitation on14day was also increased compared withAd-GFP group. miR-30a reduced the expression of ALP on BMP9-inducedosteogenic differentiation.The expression of OPN and calcium precipitationwere also inhibited by miR-30a,but had no effect on proliferation ofC3H10T1/2cells.Conclusion: BMP9-induced osteogenic differentiation is partiallyinhibited by miR-30a in mesenchymal stem cell line C3H10T1/2.
