Objective To observe the expression of bone morphogenetic protein 2(BMP-2) and bone morphogenetic protein 7(BMP-7) during corneal wound healing in rats, and to discuss the effects of BMP-2 and BMP-7 on corneal wound healing in rats.Methods Forty-two male Wistar rats were divided into seven groups, six rats in each group. The experimental groups were made into cornea model by needle puncture at their corneas of right eyes. The corneas were surgically removed at six different time points (6h, 1d, 3d, 5d, 7d, 14d) after injury. The localization and expression of BMP-2 and BMP-7 was determined by immunohistochemical methods and computer image analysis.Results The expression of BMP-2 of experimental groups reached the maximum on the 1st day in the injured corneal epithelium after operation and significant difference (P<0.05) . Then it decreased gradually on the 3rd day and 5th day. And it reached the normal level on the 14th day. The expression of BMP-7 of experimental groups decreased gradually on the 1st day and 3rd day in the injured corneal epithelium after operation, and it reached the lowest at the 5th day and significant difference (P<0.05). It increased gradually after 7 day afterward, and it reached the normal level on the 14th day.Conclusion BMP-2 and BMP-7 were expressed on rats' corneas and may play a role on corneal wound healing in rats after injury. BMP-2 was significant increased during early period after injury, however, BMP-7 may serve as negative growth regulator in the injured cornea.
