The morphological changes of EMT induced by TGF-β1in HeLa cells were observed under phase-contrast microscopy;The morphlogicalchanges of EMT induced by TGF-β1in HeLa cells caused by CAS in differentconcentrations were assesed by HE stain;Migration and invasion(with matrigel) assaywas conducted in the transwell chamber;The expressions of E-cadherin、N-cadherinand Twist were analyzed by Western blot.Results The HeLa cells were transformed by the polygonal epithelial cellsmorphology into the interstitial cells morphology after it were treated with TGF-β1at1.0、5.0and10.0ng/mL for72h, expressed as cells become spindle cells and theintercellular space was widened in concentration-dependent manner. the expression ofepithelial maker protein E-cadherin up-regulated and the mesenchymal maker proteinN-cadherin down-regulated after Hela cells were treated by TGF-β1at10.0ng/mL.CAS can inhibit the Hela cells in mesenchymal morphology (fusiformfibroblast-like) transformation after the cells were induced by TGF-β1at10.0ng/mL,and down-regulated expression of N-cadherin and up-regulated expressionof E-cadherin with the concentration-dependent manner. Compared with the controlgroup which was treated by TGF-β1at10.0ng/mL,the ability of migration andinvasion of HeLa cells was significantly reduced after the cells were treated by CASfor72h, while the expression of Twist down, in a concentration-dependent manner. Conclusion1.
