Study Of Inhibitory Effect Of Casticin On Invasion And EMT Of Lung Cancer Stem-like Cells From Human Small Cell Lung Cancer Cell Line H446

Objective To investigate the inhibitory effect of Casticin on invasion and EMT of lung cancer stem-like cells from human small cell lung cancer cell line NCI-H446, and preliminarily explore the mechanisms underlying that. To provide new train of thought and theoretical basis for drug treatment of small cell lung cancer.Methods Human lung cancer NCI-H446 cell line was cultured in vitro. LCSLCs enriched and obtained by suspension culture with stem cell-conditioned medium. Flow cytometry(FCM) with PE-CD133 antibody detected expression of CD133+ of LCSLCs. Ability of invasion and migration between LCSLCs with Parental cells measured by Transwell assay.The effect of expression of E-cadherin,N-cadherim,Twist and Snail of LCSLCs induced by CAS were analyzed using Western Blot.Results 1.LCSLCs showed typical three-dimensional spheroid with non-adherent and high expression of CD133+ in suspension culture with stem cell-conditioned medium.2.Compared with the human small cell lung cancer cell line NCI-H446,LCSLCs had a strong ability of invasion and migration in vitro.3. CAS could significantly inhibit the invasion and migration of LCSLCs in vitro.4. CAS could significantly up-regulation expression of E-cadherin of LCSLCs, could significantly down-regulation expression of N-cadherim,Twist and Snail of LCSLCs.Conclusion 1. Compared with the human small cell lung cancer cell line NCI-H446,LCSLCs had a strong ability of invasion and migration in vitro.2. CAS could significantly inhibit the ability of invasion and migration and EMT in LCSLCs in vitro.3.
