The Study Of1-Methyl-Hydantoin Inhibits The Growth Hormone Of Rats

Objective:Exploring1-Methyl-Hydantoin whether can lower levels of growth hormonewhether has the functions of somatostatin analogues，This exploration and researchprovides experimental foundation and theoretical basis for developing of non-peptidesomatostatin.Methods:Experimental subject:80female SD rats,weight230-250g.Divide into groups:80female SD rats were randomly divided into the vein groups (V1-V5groups) andabdominal cavity groups(P1-P5groups) according to the different administrationroute，altogether10groups.Each group set up blank control group (V1, P1group),model control group (V2group, P2group), the experimental group (V3-V5group andP3-P5group),8SD rats in each group.Processing: Blank control group (V1group, P1group) give0.5ml normal saline by intraperitoneal injection.Molding:model controlgroup (V2group, P2group)give10mg/ml phenobarbital for molding by intraperitonealinjection0.5ml.The experimental group (V3-V5group, P3-P5group) give10mg/mlphenobarbital for molding by intraperitoneal injection0.5ml at first, after10minutes,each experimental group inject with drugs according with different method and doseat second:V3group and P3group inject0.5ml of1-Methyl-Hydantoin10mg,V4groupand P4group inject0.5ml of1-Methyl-Hydantoin1mg,V5group and P5group inject0.5ml of1-Methyl-Hydantoin0.1mg,Blank control group (V1group, P1group) andmolding:model control group (V2group, P2group)inject PBS o.5ml.Sampling:Allgroups are get blood through tail vein,in20minutes,30minutes and40minutes,afterin injection of normal saline or phenobarbital for molding. The blood werecentrifuged and extracted the upper serum.Test: according to the instructions of rats growth hormone ELISA kit test serum growth hormone levels, contrasting each groupof growth hormone level.Results:1.The growth hormone levels in model control group higher than the blank group(P<0.01);2.Each experimental groups of growth hormone levels lower than the model controlgroup (P<0.01);3.Comparing among vein experimental groups（V3-V5groups）:the growth hormonein V3group is the lowest by vein injection1-Methyl-Hydantoin10mg(P<0.05),andthe V3group has the most obvious effect about inhibiting SD rats of growth hormone.4.Comparing among abdominal cavity experimental group（sP3-P5groups）:the growthhormone in P3group is the lowest by intraperitoneal injection1-Methyl-Hydantoin10mg(P<0.05),and the P3group has the most obvious effect about inhibiting SD ratsof growth hormone.5.The same dose of intravenous experimental groups are compared with abdominalcavity experimental groups about the growth hormone:the growth hormone levels ofvein experimental groups is lower than abdominal cavity experimental groups,thegrowth hormone in V3group lower than P3group (P<0.05) after administration in10minutes and20minutes (after molding in20minutes and30minutes), the growthhormone in V4group lower than P4group (P<0.05) after administration in10minutes,20minutes and30minutes (after molding in20minutes,30minutes and40minutes).6. The growth hormone in model control groups (V2group, P2group)is the highestafter molding in30minutes,the growth hormone in experimental group(V3-V4groupand P3-P5group) is the lowest after molding in30minutes(P<0.05)，the growthhormone in V3group and P3group after administration in20minutes (after molding30minutes) lower than the growth hormone after administration in10minutes and30minutes(after molding20minutes and40minutes)(P<0.05),he growth hormone in P4 group and P5group after administration in20minutes (after molding30minutes)lower than the growth hormone after administration in30minutes(after molding40minutes)(P<0.05).Conclusions:1.1-Methyl-Hydantoin has the effect of reducing growth hormone levels on rats, and itshows dose-dependenceit between0.1mg~10mg,it may be a new type of non peptidesomatostatin。2.1-Methyl-Hydantoin about inhibition effect of growth hormone on rats obtain thebest effect in20minutes after administration。3.1-Methyl-Hydantoin about inhibition effect of growth hormone on rats showed thevein injection approach was better than that of the abdominal cavity.
