Objective:Male Sprague Dawley rats were were subjected to aMiddle Cerebral Arteria Occlusion (MCAO), To evaluate the function andthe probably mechanism of resveratrol in cerebral I/R-induced injury.Methods: male Sprague-Dawley rats were randomly assigned to fourgroups: the sham-operation group (received a single intraperitoneal injectionof0.9%NaCl), the I/R group (received an intraperitoneal injection of0.9%NaCl at10minutes before operation as well as0and20h after reperfusion),and the control group (EDA treatment group)and treatment group(REStreatment group)(I/R+E and I/R+R groups, respectivelyreceived anintraperitoneal injection of10mg/kg edaravone or20mg/kg resveratrol at10minutes before operation as well as0and20h after reperfusion). Infarctvolume was evaluated by2,3,5-tripenyltetrazolium chloride staining, brainedema was evaluated by water content in reperfused brain, andmalondialdehyde (MDA) was detected by the thibabituric acid method.Superoxide dismutase (SOD) levels were detected using the TotalSuperoxide Dismutase Assay Kit with WST-1. Inducible nitric oxidesynthase (iNOS) levels in the hippocampus and cortex were evaluated by enzyme-linked immunosorbent assay, and aquaporin4(AQP4) expressionwas detected by immunohistochemical staining and western blot analysis.results:our data show that resveratrol decreased infarct volume andincidence of brain edema and decreased neurological deficit score. Theseoutcomes were accompanied by decreased MDA, decreased iNOS level,increased SOD level, and decreased AQP4expression in cerebral I/R injury.Conclusion: resveratrol protects against cerebral I/R injury by amelioratingoxidative stress and decreasing AQP4expression.
