ObjectiveTo investigate the effect of Galectin-7in the invasion、metastasis andearly recurrence of esophageal squamous carcinomaMethords(1) Used immunohistochemical SABC method to detect the express-ion of Galectin-7and MMP-9in the esophageal squamous cell carcinomaspecimens of50patients and38noncancerous specimens operated betweenJune2012and March2013,and to analyze the clinical pathological signify-cance of Galectin-7and MMP-9in esophageal squamous cell carcinoma.(2) Used western blot to detected the expression of Galectin-7in theconcentrated cell culture supernatant. Eca-109cell incubated with humanrecombinant Galectin-7and β-lactose in different concentrations. The experssion of the mateix metalloproteinase-9(MMP-9)、p38、p-p38in Eca-109cell analyzed by western blot and real-time quantitative PCR. The scratchtest and MTT were used to detect the impact of Galectin-7on the invadingand the proliferation of Eca-109cell.Result(1) The results demonstrated that in ESCC,the positive expression ofGalectin-7was extraordinary higher than in noncancerous specimens (χ2= 33.250,P=0.000).The positive expression of MMP-9was higher in ESCCthan in noncancerous specimens(χ2=25.539,P=0.000).The expression pa-ttern of Galectin-7was associated with clinical stage(P=0.000)、invasivedepths(P=0.000) and lymph node metastasis(P=0.012),but not withhistological differentiation、age and gender(P>0.05). The expression pat-tern of MMP-9was associated with histological differentiation(P=0.021)、clinical stage(P=0.000)、invasive depths(P=0.000)and lymph node me-tastasis(P=0.028),but not with age and gender(P>0.05).The result of me-an optical density was consistent with the expression pattern,the MOD ofGalectin-7was associated with clinical stage(P=0.000)、invasive depths(P=0.002) and lymph node metastasis(P=0.030),but not with histolo-gical differentiation、age and gender(P>0.05). the MOD of MMP-9wasassociated with histological differentiation (P=0.030)、 clinical stage(P=0.000)、invasive depths(P=0.000) and lymph node metastasis(P=0.000),but not with age and gender(P>0.05).(2) The result of western blot had testified the existence of Galectin--7in the cell culture supernatant. The results of qRT-PCR and Western blotclearly demonstrated that the expression of MMP-9was induced by galecti-n-7.And we also found compared with the control group,the expression ofp-p38in experiment group was in a higher level. In addition,the result ofscratch test showed that Galectin-7could improve the ability of invading ofEca-109.However,MTT showed Galectin-7had no effect on the prolifera- tion of Eca-109cell line.ConclusionsWe suggest that Galectin-7and MMP-9might play important roles inthe invasion and metastasis of esophageal squamous cell carcinoma. Foresophageal squamous cell carcinoma prognosis, combined detection of Galectin-7and MMP-9in ESCC can be better at predicting the recurrence andmetastasis of ESCC. In Eca-109cell line,Galectin-7was secreted to theextracellular,and induced the expression of MMP-9through activating thep38MAPK pathway by binding the specificity glycosyl receptor in the cellmembrane.
