Object:Through the study of colorectal cancer liver metastasis-associated proteins MMP2, MMP9and TIMP2, This paper aims to determine the influence of WD-3on their expressions and presents a preliminary exploration of their mechanisms; This paper also investigates the influences of WD-3-combined FOLFOX6chemotherapy on physical conditions, clinical symptoms, recent objective responses, tumor marker level, quality of life of patients, as well as the side effects of chemotherapy, when it is used to treat colorectal cancer liver metastasis.Methods:Spleen retention method is adopted to establish the animal model of transplanted colorectal liver metastases in BALB/c mouse spleen, and the model group, low-dose, middle-dose and high-dose groups of traditional Chinese medicine, Chinese and western medicine group and western medicine group are set up. Serum CEA level is determined by ELISA method; MMP2, MMP9and TIMP2’s expressions in hepatic carcinoma focus are also detected by S-P method; Changes in survival period, ascites inhibition rate, the number of metastatic nodules, and cancer weight/liver weight between various groups are compared; Serum CEA levels and MMP2, MMP9and TIMP2’s protein expressions are also compared among various groups. Based on the statistical analysis of experimental data, the influences of WD-3on the expressions of cancer liver metastasis-associated proteins are discussed to present a preliminary exploration of its mechanism; The40cases are divided into standard chemotherapy control group and WD-3+chemotherapy group. A randomized clinical trial design method is adopted to observe the changes in physical conditions, the improvement of clinical symptoms, recent objective responses, tumor marker levels, quality of life and toxicity of chemotherapy. In addition, a statistical analysis of the differences in the above indicators between the two groups are presented.Results:This experiment research finds out that compared with the model set, the traditionalChinese medicine’s low dose group, medium dose group and high dose group, the Chinese and western medicine group and the western medicine group are all capable of prolonging mice’s lifetime, decreasing the average volume of ascites and ascites’inhibition ratio, reducing the content of serum CEA, the liver metastatic nodule number and carcinoma weight or liver weight. The most noteworthy is that the Chinese and western medicine group is the most distinguished one in all the treatment groups (P<0.05), while there exists no obvious differences among the other treatment groups (P>0.05). Compared with other groups, the Chinese and western medicine group and the model set are endowed with more statistical significance (P<0.05) when it comes to reducing the expressions of metastases MMP2and MMP9protein, and increasing the expression of TIMP2. Besides, the comparation between groups demonstrates no evident statistical discrepancy (P>0.05). In terms of clinical research, the treatment groups are superior to the control groups in the aspects of improving physical power, enhancing life quality, remitting clinical symptoms, reducing the level of tumor markers and alleviating toxic and side effects of chemotherapy (P <0.05).Conclusion:WD-3can inhibit the colorectal cancer which in mice liver metastases significantly. We guess it may be through reduce liver metastases MMP2, MMP9protein and raise TIMP2protein expression to realize. The patients with advanced cancer were treated with WD-3and FOLFOX6regimen, their physical situation and life quality all improve dramatically, Meanwhile, the clinic symptoms of patients have improved noticeably,also,it can reduced tumor marker levels and the toxic side reaction of chemotherapy to some degree.
