Object:TO research on WD-3of the expression of the influence of VEGF、CK20and nm23-H1in liver metastases from colorectal carcinomace, and the preliminary discusses the mechanism.To observe the clinical effect of WD-3on the physical strength, clinical symptoms, new future curative effect, serum tumor sign, chemotherapeutic toxicity in liver metastases from colorectal carcinomace.Methods:Establish a liver metastases animal mode with Colonic cancer cells in mice inoculated spleen (CT26), set up the model group:the Chinese medicine is low, medium and high dose group、the Chinese medicine group、the traditional and western medicine group and the western medicine group. By ELISA test serum CEA content, S-P method to detect VEGF, nm23-H1and CK20focal liver cancer in the protein expression, and the comparison group in terms of survival, ascites inhibition rate, liver metastasis nodules number and heavy/liver cancer of the heavy, serum CEA content changes and VEGF, nm23-Hl and CK20protein expression, statistical analysis of experimental data, discusses WD-3of large liver metastasis associated genes influence of protein expression, and the preliminary discusses the mechanism.45patients with liver metastasis in colorectal cancer patients were randomly divided into standard chemotherapy group and WD-3united chemotherapy group (namely the treatment group), two groups of patients after treatment observation physical condition, the clinical symptoms improved situation and recent clinical curative effect, tumor markers level, quality of life and chemotherapy the reaction and toxicity of the change, and statistical analysis of two groups in the above indexes of the differences.Results:Through the experiment study found that, compared with the model group, Chinese traditional medicine low, medium and high dose group of traditional and western medicine group, the group can prolong the mice are survival time, reduce the average abdomen water and ascites inhibition rate, reduce serum CEA content, reduce liver metastases and reducing the number of nodules cancer heavy/liver heavy, which group is obviously superior to traditional model group (P﹤0.01), and is better than the other treatment group (P﹤0.05), the other no obvious difference between the treatment group (P﹥0.05); Traditional than any other group of metastases nm23-H1expression of the rise, reduce metastases VEGF, CK20expression, group compared with model was statistically significant (P﹤0.05). Other treatment group may also increase metastases nm23-H1expression, reduce metastases VEGF, CK20expression, and model group there were no statistically significant (P﹥0.05), group is also obvious difference between (P﹥0.05). Clinical research, the treatment group in improving physical condition, alleviate clinical symptoms and reduce tumor marker, improving quality of life, and reduce the adverse reaction of chemotherapy, compared with controls, the difference was statistically significant (P﹤0.05). But after treatment, the two groups were compared to the clinical curative effect, disease control rates (CR+PR+SD) were95.70%,86.40%, and also have no significant difference (P﹤0.05).Conclusion:WD-3can clearly inhibit the colorectal cancer in mice liver metastases,its mechanism may be through the lower liver metastases VEGF, CK20protein expression, higher nm23-H1protein expression to realize. WD-3united FOLFOX4scheme for colorectal cancer patients with liver metastases can improve physical condition, alleviate clinical symptoms and reduce tumor markers level, and improve the living quality and reduce the adverse reaction of chemotherapy.
