Investigation The Mechanism On The Neurovascular Regeneration With Tongnao Decoction After Ischemic Stroke In MCAO

Object To investigate the mechanism on the neurovascular regeneration with Tongnao decoction after ischemic stroke in MCAO in order to provide the laboratory basis on clinical treatment.Methods Select24adult male rats for preparation which were divided into Model group16and Sham group8.Then we made the16rats to establish rat model of cerebral infarction by throed embolism method and divided into treatment group and control group in half points.After24h,Tongnao decoction or isotonic normal saline were given by intraperitoneal injection.we took the neurological deficit score at2h,1d,7d,14d after the success of modeling.Rat cerebral infarct volume was measured by using hematoxylin-Iraq red staining at14d.Taking damage surrounding parts of the lateral ventricles and brain tissues using,real-time PCR method was used to detect the genetic variation about VEGF, bFGF, SDF-1/CXCR4amount.Results Neurological score in the treatment group was significantly lower than the control group after7days (P<0.05), no significant change in the sham group; differences in infarct volume between control group and the treatment group were statistically significant (P<0.05); RT-PCR results showed that the treatment group after14days of infarction in VEGF, bFGF, SDF-1, CXCR4gene was significantly higher compared with the control group, the difference was statistically significant (P<0.05).Conclusions Tongnao decoction can improve cerebral ischemic neurological recovery and reduce infarct volume in rats.It may be related with up-regulating VEGF, bFGF, SDF-1, CXCR4gene that can promote the cerebral vascular regeneration.This result can give us a laboratory of theoretical support for using Tongnao decoction to treat ischemic stroke in the clinic.
