Objective:To observe the Puerarin’effect on the neurological function scores and to investigate the effect of the expression of brain derived neurotrophic factor in rats with middle cerebral artery occlusion.Further to explore the cerebral protective effect of Puerarin.Methods:SD rats’ middle cerebral artery were blocked by the plugging wire,and the successful rats were used as research object.These rats were divided into the sham operation group, the model group, the low dose group and the high dose group, and each group had 24 rats.Each group was randomly divided into 1d,3d,7d,14d of the 4 time points, each time point had 6 rats.The sham operation group and the model group were given intraperitoneal injection of normal saline, and the experimental group were given intraperitoneal injection of Puerarin.In the first day, third day, seventh day and fourteenth day, according to the Neurological Severity Scores we observed the rats’nerve function scores.And in the fourteenth day we taked the rat brain slices and used HE to stain, then calculated the infarct size.Real-time PCR method was used to detect the expression of BDNF in the ischemic brain tissue of rats at each time point.Results:(l)The neurological function scores of the low dose group were obvious reduced in 3d,7d and 14d, the result had significance(P<0.05). The high dose group’difference was statistically significant in 7d and 14d. (2)Compared with the model group, the cerebral infarct area was obvious decreased in the low and high dose group,the result had significance(P<0.05). (3)Compared with the model group, the expression of BDNF in the brain tissue of the treatment group was significantly increased, especially in the 1 to 3 days after the infarction, the comparison was significant(P<0.05).Conclusion:(1)The low and high dose Puerarin can significantly improve the nerve function in MCAO model rats.(2)The low and high dose Puerarin can significantly reduce the infarct size of MCAO model rats after cerebral infarction..(3)Compared with the control group, the expression of BDNF in the brain tissue of the low and high dose group was significantly increased, especially in the 1 to 3 days after the infarction.It was showed that the use of Puerarin after cerebral infarction had protective effect on the nerve and can promote nerve regeneration to repair the damaged brain tissue.
