| Objective:Observe the effect of acupuncture on the fasting blood glucose of patients with type2diabetes. Further more, observe the effect of acupuncture on islet beta cell apoptosis in type2diabetic rats from the mitochondrial pathway. To study the cellular and molecular mechanism of acupuncture inhibition of beta cell apoptosis in type2diabetes.Methods:(1) The experimental study:The diabetic rats which were successful modeled were randomly divided into model group, acupuncture treatment group (referred to as acupuncture group), intradermal needle subcutaneous shallow needling group (referred to as intradermal needle group), western medicine of rosiglitazone treatment control group (referred to as the medicine group). Syngeneic non-diabetic SD rats were used as normal control group (normal group).Rats in the acupuncture group select the acupoints of neiting zusanli and yushu, Combined with Electro acupuncture at the same time. Treat once a day, five times a week, and select the acupoints of left and right side in turn. Rats in the intradermal needle group selects the same acupoints as the acupuncture group, and pierce the skin only with the intradermal needle. Rats in the medicine group taking rosiglitazone orally4mg/kg daily morning at the fixed time. Rats in the model group and the normal group don’t take any treatment, grasping and fixed only. Continue treatment for4weeks, and then take the blood and pancreatic tissue from the rats of each group. Test fasting plasma glucose(FPG), blood lipid, the pro apoptotic protein which called Bax, the anti apoptotic protein which called Bcl-2, caspase, and cytochrome C in the cytoplasm.(2) The clinical study:15cases of patients with type2diabetes were treated by Acupuncture. Taking the acupoint of Hegu, Quchi, Yishu, Zhongwan, Zusanli, Neiting. Reserving the needles for30minites, and needle them every10minites. Treat every other day, three times a week, and for12weeks. After the treatment, test the patients’FPG, TC.Results:(1) The experimental study:①The changes of FPGand blood lipid of rats in each group after the treatment:FPG of the acupuncture group and the intradermal needle group is under the one of model group((P<0.05). The total cholesterol(TC) of the acupuncture group and intradermal needle group is under the one of model group (P<0.01).②Bax, Bcl-2and caspase determination:Bax of the normal group,medicine group, and intradermal needle group is under the one of model group(P<0.05). Bcl-2of the normal group, acupuncture group, and intradermal needle group is higher than the one of model group (P<0.01),and medicine group is high than model group (P<0.05),and intradermal needle group is higher than medicine group (P<0.05) Caspase-3,6of the normal group and intradermal needle group is under the one of model group(P0.05). Caspase-9of the normal group and intraderma needle group is under the one of model group(P<0.05).③cyt-c determination::cyt-c of the normal group is under the one of model group(P<0.01).(2) The clinical study:FPQ TC after treatment were lower than those before therapy (P<0.01).Conclusion:(1) The experimental study:①Acupuncture and intradermal needle can effectively reduce the fasting blood glucose and blood lipid. Effect is better than that of model group. Wherein, intradermal needle group is better than the medicine group.②Intradermal needle, acupuncture, western medicine rosiglitazone can suppressed the pro apoptotic protein, protect the anti apoptotic proteins, and inhibit the activation of caspase, thereby inhibiting the apoptosis of pancreatic beta cells. Wherein, intradermal needle is better than acupuncture and rosiglitazone.③Acupuncture, intradermal needle and rosiglitazone can reduce cyt-c content in mitochondria. Wherein, intradermal needle is better than acupuncture and rosiglitazone. This proves the intradermal needle can effectively inhibit the apoptosis of pancreatic beta cells.(2)The clinical study:Acupuncture therapy can effectively reduce the FPQ TC of patients with T2DM. |
