| Objective:To investigate the frequency distribution of PCSK9gene I474V(rs652556) polymorphisms and their relationship in patients with ischemic stroke(IS) in Xinjiang Uygurs and Hans populations and the main risk factors of ischemic stroke.Methods:The I474V polymorphism was identified by polymerase chain-reaction restriction fragment length polymorphism (PCR-RFLP) in407patients with IS (including219Hans and188Uygurs) and425health controls (including255Hans and170Uygurs),Some specimens were sequenced,and ischemic stroke’s risk factors were analyzed. Results:(1)There were significant association with body mass index,smoking history,drinking history, hypertension,hyperlipidemia,diabetes,mellitus and ischemic stroke(P<0.05); Multivariate conditional Logistic regression analysis of ischemic strok risk factors showed that there were significant association with hypertension(odds ratio,5.605, P<0.001), diabetes mellitus(odds ratio,3.026, P<0.001), smoking(odds ratio,2.781, P<0.001), alcohol (odds ratio,2.770, P<0.001),hyperlipidemia(odds ratio,2.456, P<0.001), body mass index (odds ratio,1.703, P=0.006)and ischemic stroke.(2)Between IS group and control group,the â…¡ and IV genotypes had no statistical significance differences in the levels of triglycerides(TG)ã€high-density lipoprotein cholesterol(HDL-C)(P>0.05);total cholesterol(TC)ã€low-density lipoprotein cholesterol(LDL-C) levels had statistical significance (P<0.05);Between IS and control groups,â…¡ genotype of TC,LDL-C,HDL-C levels were statistical difference (P<0.05);In the IS group IV genotype TC%LDL-C levels were significantly higher than the control group,the difference was statistically significant (P<0.05).(3)There was statistical difference in the genotype distribution between IS and control group(9.5%,4.5%;χ2=16.09, P=0.00).And the distribution of allele frequency is the similarity(18.9%,8.9%;χ2=17.38, P=0.00).(4)The gene frequency and genotypes of I474V showed significant difference in both the Xinjiang Uygurs and Hans goups(27.7%ã€11.4%,χ2=17.40, P=0.00;12.9%,6.9%,χ2=5.57, P=0.018).(5)There was statistical significant difference the genotype distribution and allele frenquency distribution between IS group and the control group in the Xinjiang Uygurs (27.7%ã€12.9%,χ2=11.79, P=0.001;13.8%ã€6.5%,χ2=10.44, P=0.001);But there was no statistical significance in the Hans (P>0.05).Conclusion:Hypertension, diabetes mellitus, hyperlipidemia, drinking history,body mass index are the major risk factors for ischemic stroke;â…¡ and â…£ genotypes are dominant in the rs652556loci polymorphism of PCSK9gene.The genotype of PCSK9gene rs652556loci polymorphism is correlated with serum increasing levels of TC and LDL-C,We have found rs652556SNPs is associate with cerebral ischemic stroke course in xinjiang region,There are statistical significance in the difference of the genotype rs652556distribution between Uygurs and Hans goups, rs652556SNPs have a relationship to the occurrence of IS in the Xinjiang uygurs,IV may be susceptible genetypes and V may be a genetic susceptible alleles of the Xinjiang uygurs. |
